PURIFICATION METHOD: Not applicable

SPECIFICITY: Hodgkin and Reed-Sternberg cells of all histological sub-types of Hodgkin's lymphomas. The CD30 antigen consists of two polypeptide chains, mol.wt. 105 and 120 kDa


APPLICATIONS:

* Detection of Reed-Sternberg cells and especially mononuclear Hodgkin's cells.

* Suitable on cryostat sections and cell smears only.

PRESENTATION: Tissue culture supernatant dialysed against 0.05M Tris/HCl, pH 7.2 containing sodium azide. Sufficient for 200 tests (1ml)

STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -20C. After thawing they should then be used as a stock solution for up to 60 days. Dilute working solutions are best prepared on the day of use.

Activity: Reacts with a 120Kd type I transmembrane glycoprotein expressed on in vitro stimulated T and B cells. CD30 is an activation marker which was initially identified on Reed- Sternberg cells and is also expressed on a few extrafollicular T and B cells located at the rim of germinal centers. It is a cytokine receptor which belongs to the nerve growth factor receptor/tumor necrosis factor receptor superfamily. CD30 interaction with CD30 Ligand (CD30L) can mediate signals for proliferation, apotosis and cytotoxicity of lymphoid cells. Clone BerH8 is suitable for staining formalin fixed paraffin embedded tissue sections without pretreatment.

Activity: Reacts with human 30. Recognizes CD30 molecule of about 120kda.

Application: flow cytometry

-500K cultured CEM human tumor cells were washed and incubated 45minutes on ice with 80ul of anti-CD30 antibody at 20ug/ml. Cells were washed twice and incubated with secondary goat anti-mouse IgG:FITC, after which they were washed three times, fixed and analyzed using a BD FACstar plus. Cells stained positive with a mean shift of 0.39log10 fluorescent units when compared with a mouse IgG2B negatove control. -titers must be determined for each application
