Reactivity: Reacts with the murine CD40 molecule, a 40-50 kDa glycoprotein
expressed on B cells, follicular dendritic cells, thymic epithelium and
interdigitating cells in the T cell zones of secondary lymphoid organs. The
CD40 molecule has a central role in B cell growth and differentiation:Ligation
of CD40 is reported to be an important signal in B cell survival. Furthermore,
signaling through CD40 in combination with IL-4 reportedly induces immunoglobulin
isotype switching and secretion of IgE.
-Clone 3/23 also reacts with 5-10% of T cells in the adult but not with neonatal mouse spleen. The reactivity of 3/23 with peripheral T cells has been observed in some but not all mouse strain analyzed. Ligation of CD40 with clone 3/23 antibody upregulates the expression of MHC Class II antigens, CD23 and CD86. Although this 3/23 antibody by itself is a weak B cell mitogen, it synergizes markedly with mitogenic anti-IgM or anti-D mab or IL-4 to promote B cell proliferation. IN addition, co stimulation with 3/23 and IL- 4 rescues B cells from apoptosis by extensive cross linking of surface Igm or IgD.

-Plate bound or soluble HM40-3 antibody stimulates spleen cells to proliferate in vitro. This antibody also induces spleen B cells to express the co-stimulatory molecule CD86. Clone HM40-3 has been demonstrated to inhibit the binding of soluble gp39, the CD40 Ligand to soluble CD40 and to cell surface CD40.
-Also cross reacts with simialr opulations of Lewis, Sprague-Dawley and LOU rat leukocytes.
