RDI Divison of Fitzgerald Industries Intl  offers a wide line of  antibodies. Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Rabbit anti-Dopamine D2 receptor     cat#RDI-D2Rabr

$406.00/100ul neat sera with 0.05% sodium azide

-made against D2R sequence that lies in the amino terminus just before the first transmembrane domain (sequence does not show any similarity to other members of G-coupled receptors)

-recommended for Light microscopy and immunoblotting

-reactivity with mouse, rat, human, lobster, squid and chicken

Rabbit anti-Dopamine D2L Receptor (Rat/Mouse

Presentation: 100ug affinity pure antibody (lyophilized from 1mg/ml 100mM Tris pH7.5 with 0.2% BSA and 0.05% sodium azide).

Reconstitution: Add at least 100ul sterile DI water, let set at least 30 minutes at 4 DEG C. Gently mix well. If desired, centrifuge 1-2 minutes at 1000 rpm to concentrate material in vial. Stock=1mg/ml. Recommend making any further dilutions into buffers with 0.1-1% BSA or other carrier and suitable preservative as 0.05% sodium azide.

Purification: peptide affinity purified.

Antigen: a 27aa peptide sequence from the rat D2 receptor long form. No significant homoloogy is seen with the other dopamine receptors.

Reconstitution: All peptide/antibody received in solution or reconstituted from Lyophilized vials should be stored frozen at -20oC or below in suitable aliquots. It is not recommended to store diluted solutions. Avoid repeated freeze and thaw.

Specificity: the 27 aa rat D2Rlr peptide sequence shows homology : 96% human/bovine/dog    64% Xenopus

USE: Western Blotting (1-10ug/ml with chemiluminescence detection) -must optomize for each application

ELISA (1:1K-1:10K; using 1ug/ml of control peptide coated to plate).

Immunoprecipitations . not tested

Histochemistry & Immunofluorescence: not tested titer from 10ug/ml using overnight incubation and ABC/Dab/enhanced detection.

Generally titer antibody to maximum usable dilution and then pretreat antibody with 10X weight of peptide for 2-3 hours RT or overnight 4 DEG C, then proceed with staining.

Rabbit anti-Dopamine D2 Receptor (Rat/Human/Bovine+)

Presentation: 100ul neat sera with 0.05% sodium azide).

-at first use, tighten cap, centrifuge 1-2 minutes at 1000 rpm to concentrate material in vial. Recommend making any dilutions in buffers with 0.1-1% BSA and 0.05% sodium azide

Antigen: An Amino terminal, 11 amino acid peptide sequence (24-34AA) near or at the ligand binding domain of rat D2R. This peptide sequence is right before the first membrane spanning domain. The sequence does not show any similarity to the other members of G- coupled receptors. The preptide was coupled to KLH.

Reconstitution: All peptide/antibody received in solution or reconstituted from Lyophilized vials should be stored frozen at -20oC or below in suitable aliquots. It is not recommended to store diluted solutions. Avoid repeated freeze and thaw.

Specificity: the 11 aa rat D2Dr peptide sequence shows 100% homology with mouse, 90% human and bovine and 72% with dog D2DR receptors. The antibody has been shown to cross react with human (CNS and peripheral), Lobster, squid and crawfish.

USE: Western Blotting (1:500-1:1K of neat sera using Chemiluminescence technique). -must optomize for each application -it may be necessary to load 100-200ug of purified striatal or other tissue membranes to visualize the D2R bands. Reactive with both native and denatured D2R bands.

ELISA (1:1K-1:10K; using 1ug/ml of control peptide coated to plate).

Histochemistry & Immunofluorescence: 1:250 - 1:750 dilution using overnight incubation and enhanced detection.

Other:flow cytometry with D2R in fibroblast transfected with rat D2R cDNA, inhibition assays

-sequence: NH2-C-A-A-R-R-A-Q-E-L-E-M-E-COOH

Reconstitution: Reconstitute the lyophilized antibody with 100ul of PBS containing 10mg/ml BSA. For further dilutions, use a similar BSA containing Buffer.

Specificity: Recognizes the dopamine D2S and D2L receptor from human and rat (primarily the D2L form)

USE: Western Blotting (1:800 of neat sera using Chemiluminescence technique).

-detects bands at approx 48 & 51kDa band when using whole rat brain homogenates

-NOTE: FOR BLOCKING use only 4% goat serum or evaporated goat's milk.

Histochemistry :1:5000 demonstrated staining of specific cells in various regions of rat brain sections. Such staining includes plasma membrane in the cortex, caudate putamen, globus pallidus, thalmic nucleus, supra optic nucleus and the dentate gyrus.

-Optimum titers must be established for each use

Reconstitution: Reconstitute the lyophilized antibody with 100ul of PBS containing 10mg/ml BSA. For further dilutions, use a similar BSA containing Buffer.

Specificity: Recognizes the dopamine D2S receptor from human and rat

USE: Western Blotting (1:800 of neat sera using Chemiluminescence technique).

-detects bands at approx 48 band when using whole rat brain homogenates

Histochemistry :1:5000 demonstrated staining of specific cells in various regions of rat brain sections. Such staining includes plasma membrane in the cortex and caudate putamen. Staining was also observed along apical dendrites of layer 5 cortex cells. -Optimum titers must be established for each use
