PURIFICATION METHOD: gel filtration chromatography following ammonium sulphate precipitation.

SPECIFICITY: This antibody recognizes the MIC 2 (E2) antigen expressed on most human cells except those of the myeloid family. The 32kDa molecule on thymocytes and T cells is involved in rosette formation.

APPLICATIONS:

*Studies of MIC 2 antigen expression on cryostat sections and in flow cytometry.

* Antibody is positive in western blotting

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration of not less than 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml of bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at 4 DEG C. Dilute working solutions are best prepared on the day of use.

Activity: Reacts with a 32 kd sialoglycoprotein also referred to as E2 antigen, expressed on all leukocyte lineages. The E2 antigen is the MIC2 gene product and is differentially expressed during T and B lymphoid and granulocytic development, with higher densities being expressed during early development, with higher densities being expressed during early hematopoietic stages. Mature granulocytes express very little or no CD99. E2 has been shown to be involved in T-cell adhesion processes and is suggested to have a functional role in hematopoietic adhesion pathways.

Activity: Reacts with E2 molecule, a highly glycosylated 32kd single polypeptide chain. CD99R is an isoform of CD99 with restricted cell distribution. It is expressed on T, NK, and myeloid cells but not on B cells, erythrocytes, platelets or bone marrow cells. The E2 molecule plays a role in adhesion processes. Antibodies to E2 have an effect in CA++ flux on Jurkat cells, suggesting a possible role in signal transduction.
