Quality Control involves coating ELISA plates and assaying a series of panel
samples. Reactivity to the negative control must not be more than twice the
blank. Infectivity Analysis: Negative infectivity for CMV is defined as the
inability of an inactivated virus preparation to produce CPE after eighteen
(18) days of incubation on a monolayer of HF cells.
Cytomegalovirus (CMV) is considered one of the most common viral infections
in man. The virus rarely causes disease, but may persist as a latent or chronic
infection of the salivary glands, renal tubules, cervix and leukocytes. The
incidence and spectrum of the disease in newborns and immunocompromised hosts
establish the virus as an important human pathogen. CMV infections are frequent
and occasionally severe in children or adults with congenital or acquired
defects of cellular immunity, such as patients with AIDS, cancer patients,
and recipients of organ transplants. The transmission of CMV via transfused
blood may account for most CMV infections in organ transplant recipients,
neonates, and heart surgery patients. Medical conditions caused by CMV include:
interstitial pneumonia, mononucleosis (heterophile negative), congenital
defects, abortion, mental retardation, blindness and deafness.
Serological tests for the detection and quantification of antibodies to CMV
are a valuable aid in diagnosing primary infection. Serology represents the
only practical system at present for screening blood and organ donors and
recipients to prevent the transmission of latent CMV to patients at high
risk for severe CMV disease.
Cytomegalovirus Antigens are prepared by proprietary glycine extraction of
human fibroblast cells infected with cytomegalovirus, strain AD 169 (ATCC).
The CMV-M extraction procedure differs from the CMV-G procedure; it is designed
to increase the relative amount of early viral CMV antigens. Both antigen
preparations are partially purified to reduce host cell components prior
to inactivation with UV light. The CMV-G & CMV-M antigens have yielded
lot-to-lot consistency in IgG & IgM ELISA tests respectively, providing
high sensitivity and low backgrounds.
Higher antibody reactivity is achieved with lower total protein for both the CMV-G and CMV-M products as compared to two commercial products. The higher specific activity achieved through partial purification of the CMV antigens assures improved sensitivity and specificity for your assays.
