Grade: recombinant antigen is a virus-like particle (VLP) containing all
the three structural rubella virus proteins, namely c-core protein and 2
surface glycoproteins E1 and E2 but lacking genomic viral RNA.
Source: recombinant antigen is produced by continuous cell line of an animal
origin transfected by plasmid bearing the set of rubella virus structural
protein genes.
Application: Coating/capture antigen for specific IgG/IgM antibody rubella
virus detection in human sera
Purification: The recombinant cell culture is grown using alpha-MEM containing
10% FCS by roller cultivation. After reaching a continuous monolayer a tissue
culture is harvested and clarified by a slow spped centrifugation at 1000
rpm for 30 minutes. VLP are pelleted by high- speed centrifugation and further
purified by sucrose density 30-60% linear gradietn centrifugation at 25000
rpm for 2.5 hours. The VLP zone is collected, diluted ten times with PBS
and pelleted by centrifugation at 21000 rpm for 1.5 hours.
Presentation: Solution in PBS, pH7.2 containing 0.1mM PMSF
Protein conc: 1mg/ml by modified Bradford method (Bio-Rad Protein Assay Kit
II, cat> 500-0002)
Immunoreactivity: Tested in EIA and western blotting using anti-rubella Mab
clone Ru6 (see cat#RDI-TRK3R23-Ru6) and anti- Rubella virus positive human
sera.
