Sample Western blotting protocol:
We used 10 ul of the cell extract which expresses all HCV non-structural proteins with 10ul of the 2X Loading buffer (0.125 M Tris-HCl pH 6.8; 4% SDS, 20% glycerol; 10%Beta mercaptoethanol; 0.02%Bromphenol blue ). On the 10% PAGE-SDS gel. After the gel run and transferred to PVDF membrane we block with 5% Dry milk for 30 minutes and wash in 1X TBST (tris buffered saline with tween-20). The primary antibody is diluted 1 to 1:1000 (1ug/ml) incubated for 1 hour. Then we wash 3 X 10 minutes each in TBST and continue with standard protocol.
All procedures done at room temperature.
