These clones have been derived from hybridization of X63-Ag8-653 myeloma cells with spleen cells of Balb/c miceimmunized with high molecular weight (more than 300 kDa) glycoprotein from human milk-fat globule molecule.
Specificity: Clone CM1 - Peptide tandem repeat in MUC1 core.

Clone X19 - peptide repeat RPAP region
Clones M9E7, M4H2, M8C9, M10G4, M10H6: Affinity > 1 x 10 8 for all clones. Mabs bind with high efficiency with different regions of GVTSAPDTRPAPGSTAPPAHGVTSA synthetic peptide spanning the one repeat of VNTR extracellular portion of MUC1 molecule. They react with VNTR5 and VNTR20 recombinant unglycosylated fragments of MUC1 protein and underglycosilated MUC1 prepared from tumor fluids or as a result of chemical treatment of human milk MUC1. These Mabs do not recognize natural MUC1 protein isolated from human milk by affinity purification on carbohydrate epitope specific Mabs and belong to Mab cluster 1-4 according to ISOMB TD-4 classification . Non crossreactive with egg white avidin.
Clones M4H2, M2F1, M2C5, M2E10 and M2A10: A ffinity > 1 X 10 8 for all clones. Mabs bind with high efficiency with different epitopes within the VNTR tandem repeat peptide region of MUC1 molecule, which have different conformations and peptide- carbohydrate compositions. Mab M4H2 react with VNTR20 recombinant unglycosylated fragment of MUC1 protein (but not with monomeric MUC1 peptide), with deglycosilated and underglycosylated natural MUC1 (but not with completely glycosylated MUC1). Mabs M2F1, M2C5, M2E104, M3A106 efficiently recognize underglycosylated and natural MUC1 protein isolated from human milk by affinity chromatography. All these Mabs belong to Mab clusters 6 and 7 according to ISOMB TD-4 classification. Non crossreactive with normal serum proteins and human tissues of nonepithelial origin.

Background: Muc1 is a heavily O-glycosylated transmembrane protein expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and overexpressed in >90% breast carcinomas and metastases. In normal mammary gland it is expressed in apical surface of glandular epithelium. In breast cancer Muc1 is overexpressed; is underglycosylated and the apical localization is lost. Muc1 is transcribed as a larger precursor which is cleaved to form a larger mucin like subunit (265-400kDa) and a smaller subunit (14- 28kDa) noncovalently associated with each other. Transgenic Muc1 has been shown to associate with all four erbB receptors and localize with erbB1 (EGFR) in lactating glands 1 . Muc1 can act as a ligand for ICAM-1 on HUVEC cells; it can bind beta-catenin, GSK3beta and it associates with Grb2-SOS upon phosphorylation..

Background: MUc-2 glycoprotein also known as intestinal mucin related protein
antigen is an intestinal apomucin.Muc-2 glycoprotein is expressed in mucinous
tumors but not in serous tumors. Expression of MUC-2 glycoprotein is found in a
low percentage of borderline malignancies and in approx 55 percent of
carcinomas where Muc-1 glycoprotein is also expressed. muc-2 is useful for the
detection of MUC-2 glycoprotein in colonic and gastric tumors.
