Specificity: The multi-drug transport protein known as P-glycoprotein, 0170 or MDR1 gene production is directly related to the ability of selected drug resistant mammalian cell lines to proliferate in vitro in the presence of cylotoxic drugs. This plasma membrane bound protein has both cyloplasmic and extracellular domains. This antibody is known to specifically recognize the extracellular component and has definite application to the flow cytometric analysis of this molecule.
Presentation: The monoclonal is presented at a concentration of 200ug/2ml in
phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum
albumin. We recommend that each laboratory determine an optimum working
titre for use in it particular application.
Storage: For use within 1 month of purchase store at +4'C, for long term
storage aliquote antibody into small volumes and store at -20'C.

Specificity: Reacts specifically with a surface epitope of human glycoprotein.
Clone MRK16 reacts with P-Glycoprotein of human cortical adenomas and does not
react with eochromocytoma, non functioning cortical adenoma, or myolipoma of
the adrenal. MRK16 does not react with mouse P-glycoprotein. (other clones
which react with inner membrane epitopes may cross-react with heavy chains of
muscle myosin and/or type A blood).
Activity: Clone MRK16 binds to human P-Glycoprotein, a 170 kDa multi-drug resistance product that acts as an ATP-dependent efflux pump to remove drugs and other potentially harmful products from the cell interior. Research as linked multi-drug resistance in cancer cells to the over expression of P-glycoprotein. Since clone MRK16 reacts with P-glycoprotein exposed on the cell's surface, living cells can be labeled. After binding, MRK16 inhibits the function of P-glycoprotein. Glycoprotein-MRK16-protein A sepharose complex from the human adrenals possesses ATPase activity.
Uses: MRK16 shows good reactivity in FACS and immunoprecipitation. Frozen
tissue staining can be done providing that mild fixation treatment is used
(such as 4% paraformaldehyde. Acetone, paraffin, methanol and ethanol should
not be used). Not suitable for western blotting because the antibody loses its
structure in the presence of protein denaturing agents such as SDS..
For fACS use(purified antibody): 500K live cells (fixation not necessary), add
50ul of ab (10 to 100ug/ml), incubate 30 minutes on ice. Wash 3X with 10%
FCS/PBS, add 20ul FITC-anti-mouse IgG, 15 minutes on ice, Wash 3X with 10%
FCS/PBS, 500ul PBS analyze.
-for biotin conjugate: use 300K cells, use FITC:Avidin or Streptavidin-RED670
(see protocol with shipment).
For immunoprecipitation: 25-50ug/ml
For histostaining: 0.1-10ug/ml (approx)
Storage: Store at 4 Deg C. Aliquot and store at -20DEG C for long term storage. Avoid frequent freeze thaw cycles.
Precautions: For In vitro research Use Only. Not for use in or on humans or animals or for diagnostics. Sodium azide may form explosive compounds in presence of heavy metals or under acidic conditions. Flush drains with copious amounts of water to prevent buildup of explosive compounds.
