SPECIFICITY This antibody reacts with human p53 protein, wild type and mutant forms under denaturing conditions. It also reacts with mutant form under non-denaturing conditions but not with wild-type p53 protein under these conditions. In man the p53 gene is located on the small arm of chromosome 17. Alterations to the p53 tumour suppressor gene are a common feature of human malignancies, a normal function which, is to induce apoptosis after DNA damage and so its inactivation can permit the survival of cells that have sustained genetic damage. CBL 773 reacts with the human form of p53 protein at an epitope within the core region between amino acids 250 and 270 which is located between conserved domain IV and V on an exposed turn of the protein structure.

SPECIFICITY This antibody reacts with human p53 protein, wild type and mutant forms under denaturing and non-denaturing conditions. It specifically maps the n-terminus region of the p53 molecule (amino acids 56 to 65). CBL 774 has been used to study p53 protein changes in conformations and their association with different biological activities. Such conformation or changes may be important for the normal regulation of p53 function. The antibody also recognizes an additional 40 kDa band to the 53 kDa band expressed by the HOS cell line when used in Western blotting, database searches have failed to identify a protein other than p53 which contains the additional sequence recognized.
