Reactivities on Cultured All Ca-cell lines tested so far:  Particularly strong

Cell Lines (tested so far)                                     reaction with colon CA cell lines (HT-29, WiDr, SW1116)

Tumors Specifically Detected: ALL CA types (18 diff. Origins tested so far). A subset of squamous cell CA is negative. Reacts with structures of epithelial origin, not with mesothelial structures.

Pretreatment: with paraffin embedded sections, protease pretreatment is required prior to antibody application.

Working Dilution When reconstituted with 1ml distilled water dilute 1:10 With PBS, pH 7.4 for use in immunohistochemistry

Incubation Time 1 h at RT, extended time with paraffin

Storage: One year after reconstitution when stored at 2-8 DEG C, for longer storage aliquots should be kept at -20 DEG C

Note: The ESA antigen is also recognized by other antibodies including clones HEA125, GA733, 323/A3, KS1/4, 17-1A, Ber-EP4 & AUA1. The ESA gene is fully sequenced(Szala et al, 1990 and Simon, et al, 1990). This clone will react with a wide variety of epithelial tissues. The major application of the antibody is a general epithelial cell marker, but its particular use is in the distinction of adenocarcinoma from mesothelloma which does not stain in most cases ( Latza et al, 1990).

Specificity:* reacts with the human high molecular weight sialomucin, episialin (also known as PEM and EMA). The antibody recognizes the multiple protein epitope that is relatively unsensitive to glycosylation. This makes the antibody very sensitive in detection of episilan expression in different in different epithelial tissues.

SPECIFICITY This antibody recognizes a 40kDa transmembranous epithelial glycoprotein expressed on baso-lateral cell surfaces in very many epithelial tissues (but absent from mesothelial tissues). The extracellular domain has a cysteine-rich repeat and a small domain with homology to nidogen. It is a homophyllic cell-cell adhesion molecule, recently called Ep-CAM (Litvinov et al., 1994). This molecule is also detected by antibodies such as 323/A3, BER-EP4, 17-1A, AUA1, KS1/4, MOC-31, GA733, HEA 125 and others). It reacts with most epithelial cells and carcinomas.

LOT SPECIFIC DATA

PRESENTATION The monoclonal is presented at a concentration of 100µg/1ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

STORAGE For use within 1 month of purchase store at +4oC, for long term storage aliquot antibody into small volumes and store at -20oC.
