Application: -direct fluorescence (approx 80ul of 1:50 dilution/500K cells)

-Briefly, ficoll prepared human peripheral blood lymphocytes were stimulated in culture 3 days in the presence of PHA-P (5ug/ml). 500K cells were then washed and pre-incubated 5 minutes on ice with 20ul of 250ug/ml human IgG (to block nonspecific binding) and then incubated 45 minutes on ice with 80ul of product at 1:50 dilution. Cells were washed 3X and analyzed.

CLONE: BNI3.1

Activity: Reacts with the cytolytic T lymphocyte associated antigen (CTLA-4). CTLA-4 is transiently expressed on activated CD28+ T cells and binds to CD80 and CD86 present on antigen presenting cells (APC) with high avidity. This interaction appears to deliver a negative regulatory signal to the T cell. There are recent reports which indicate that CTLA-4 is also expressed on B cells when cultured with activated T cells suggesting a possible role of CTLA-4 in the regulation of B cell response.
