SPECIFICITY: The CD95 antigen, molecular weight 40-50kDa, is a cell surface
glycoprotein that was originally identified by two apoptosis inducing antibodies,
anti-APO-1 and anti-FAS. FAS is known to induce signal into cells ad anti-FAs
antibody works as an agonist for the FAS protein. CD95 is expressed on a
minority of resting T and B cells and monocytes whose viability is unaffected
by anti-CD95 antibodies. In marked contrast anti-CD95 antibodies induce apoptosis
in activated T and B cells, NK cells and thymocytes.
APPLICATIONS:
*Flow cytometric studies
*Studies of apoptosis without immobilization of the antibody, using the
EBV cell line SKW69 0.25ug/ml of CBL527 will induce 50% apoptosis in a 24
hour bioassay.
PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration
of not less than 200µg/2ml in in phosphate buffred saline containing
10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each
laboratory determine an optimum working titre for use in its particular
application.
STORAGE: Purified monoclonal antibodies are generally stable for long periods
at 4oC. However repeated warming to room temperature and re-cooling may result
in a loss of activity and hence effective working titre. It is recommended
that monoclonal antibodies are aliquotted upon receipt and then stored at
-20C. After thawing they should then be used as a stock solution for up to
30 days. Dilute working solutions are best prepared on the day of use.

Background: Fas (also known as Apo-1 and CD95) is a member of a family of
cell surface receptors which includes TNFR, NGFR, CD40, CD27, CD30 and 4-1BB.
Both the murine and human Fas genes have been cloned and share 60% similarity
in their deduced amino acid sequences. Fas is expressed in the thymus, liver,
heart, and ovary. Fas appears to be a cell surface apotosis signalling molecule
which is expressed in a number of different cell types. Abnormalities in
the Fas gene are correlated with autoimmune features in mice and unusually
high levels of lymphocyte apotosis in HIV infected humans. Genetic studies
localized the Fas gene near lpr (lymphoproliferation disease) locus on mouse
chromosome 19. Further characterization revealed that lpr is a mutation affecting
function of the Fas gene. recent cloning of the Fas ligand (FASL) revealed
that it is a member of the TNF family. It has been postulated that Fas plays
a role in T cell mediated cytotoxicity and possibly in the normal apoptotic
process that occurs during T cell development.

Activity: Reacts with the 45kD transmembrane cell surface FAS or APO-1 antigen,
designated CD95. CD95 is expressed on a variety of normal and neoplastic
cells, and is a polypeptide which plays a role in the programmed sequence
of events leading to cell death (apoptosis). the DX2 clone specifically reacts
with murine L cells, murine L1210 leukemia cells and murine P815 mastocytoma
cells transfected with human Fas cDNA but not with untransfected parental
cell lines. Cross linking with DX2 delivers an apoptotic signal indicating
that DX2 recognizes a functional epitope of CD95 antigen.

Activity: Reacts with Fas-Ligand expressed on activated T lymphocytes of
selected strains of mice (C57BL/6, C3H, MRL, NOD, NZB, NZW and SJL). It does
not react with similarly activated T blasts from BALB/c, DBA/1 or DBA/2 mice.
It also reacts with COS cells transfected with mFASL cDNA derived from C57BL/6
and C3H mice, but not with mFASL cDNA from BALB/c nor DBA/2 mice. In addition,
this clone efficiently blocks cytotoxic activity of a C3H T cell line but
not a BALB/c T cell line. Phenotypic and genotypic characterizations of mouse
FAS-Ligand reveal the existence on activated T cell lines and in spleen,
testis and eye. FASL mRNA has been demonstrated at various levels in bone
marrow, thymus, spleen, lymph node, lung, small intestine, testis and uterus.
Morever, T cell activators but not B-cell activators enhanced the expression of FasL mRNA in splenocytes; and FasL mRNA was restricted to the T cell lineage among a panel of cell lines from lymphoid tissues. FasL is not functional in mice homozygous for the gld (generalized lymphoproliferative disease) mutation; these mice cannot limit the expansion of activated lymphocytes and develop autoimmune disease. FasL is a type II membrane protein a member of the TNF/NGF family, which binds to CD95 (FAS). FasL and its counter-receptor CD95 are believed to participate in T cell development, the regulation of immune responses, and cell mediated cytotoxic mechanisms. Functional studies suggest that mFasL.2 has higher specific activity than mFasL1. It has been demonstrated that human FASL is released from the surface of transfectant cells and activated human T cells by metalloproteinases.
Application: -immunofluorescent staining in the presence of
metalloproteinase inhibitor on mFAS-L transfected and activated T cell lines
(1-4ug/million cells). NOTE:PE has been reported to bind to mouse
B cells via FCgII receptors. For immunostaining of mouse cells, this
can be prevented by preincubating cells with a CD16/CD32 antibody.

Antigen: C57BL/6 mouse FasL c DNA transfected baby hamster kidney cells
Activity: Clone MFL3 reacts with mouse Fas Ligand (Fas L), a member of the
TNF/NGF superfmaily. Fas L has been reported to be expressed by activated
mouse T cells as well as some T-cell lines and clones. Mouse FasL mRNA has
been found in spleen, thymus, lung, lymph nodes, eye and testis. Fas L is
a tyep II transmembrane protein which binds to its counter- receptor CD95
(FAS) and is speculated to participate in regulation of a variety of
immunological responses and lymphocyte development. Clone MFL3 has been reproted
to efficiently inhibit the cytotoxicity of mouse FAS-L transfected cells
against human Fas-transfected cells.
