SPECIFICITY     This antibody recognizes type II integral membrane glycoproteins (35 kDa) present on the nuclear membrane of B cells. This antigen is mainly expressed on the cell surface of resting and activated B cells. In immunoprecipitation studies, LN-2precipitates three molecules from S-labelled RJ2.2.5 cells: a weak 41-kDa band, a 35-kDa band and a strong 33 kDa band. Ln-2 has been demonstrated to react with an epitope residing within 60 amino acids of the extracytoplasmic C-terminus of the molecule.The invariant chain, CD74 is involved in MHC class ll-restricted antigen presentation and processing.

APPLICATIONS * Studies of B cells from follicular centre and mantle zone. Also labelled using Heat Induced Epitope Retrieval (HIER)* Studies on formalin fixed paraffin embedded sections*Distinguishing B cell lymphomas from T cell lymphomas. B-cell leukaemias corresponding to different stages of B-cell maturation react positively with LN-2 including pre-B and B-lineage common acute lymphoblastic leukemia (ALL), chronic lymphocytic leukemia (B-CLL) and hairy cell leukaemia. By immunohistochemistry, LN-2 has been shown to label B-cell non-Hodgkin's lymphoma on routinely processed tissue. Small lymphocytic and small cleaved lymphomas have been observed to react most strongly; however large cell lymphomas are also labelled with LN-2. Mononuclear Hodgkin cells and Reed-Sternberg cells in Hodgkin's disease have been demonstrated to stain positively, as do some rare T-cell lymphomas. Among non-lymphoid neoplasms, LN-2 has been observed to label some epithelial tumours including adenocarcinoma of the uterus, squamous cell carcinoma of the lung and transitional cell carcinoma of the bladder.Langerhans cells and cells of histiocytosis or histiocytosis X have demonstrated positively with LN-2.

PRESENTATION The monoclonal is presented at a concentration of 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

STORAGE For use within 1 month of purchase store at +4oC, for long term storage aliquot antibody into small volumes and store at -20oC.

INFORMATION: Human CD74 is expressed intracellularly and on the cell surface of MHC Class II positive cells such as B cells, monocytes, macrophages, Langerhans cells and dendritic cells. CD74 has several important functions relating to antigen presentation by MHC Class II molecules. Antibody M-B741 recognizes the CD74 molecule of about 33 kd.

STORAGE CONDITIONS: Store at 2 - 5oC . Freeze/Thawing is not recommended.

PERFORMANCE: Briefly, 500K cultured Raji cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD74 antibody at 5 m g/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC at a 1:60 dilution factor, after which they were washed three times, fixed and analyzed using a BD FACstar plus. Cells stained positive with a mean shift of 1.09 log10 fluorescent units when compared to a Mouse IgG2a negative control at a similar concentration.
