Specificity: Cell type: Activated B and T lymfocytes.   Ag.MW.: 60 kD.

Reactivity: The monoclonal antibody is directed against the CD70-antigen,which is expressed on activated B and T lymphocytes. The monoclonal antibody also reacts positive with Sternberg-Reed cells.

Applications: Detection of activated B-and T-cells.

Methods: Indirect immunofluorescence staining with analysis by FACS or fluorescence microscopy.

Clone: 2F2. This clone has been derived from hybridization of SP2/0 cells with spleen cells of BALB/c mouse immunized with irradiated Ramos cells.

Mouse isotype:The monoclonal antibody is of the mouse subclass IgG1.

Source: Hybridoma culture supernatant.

Purification: Ammoniumsulphate precipitation and ion exchange chromatagraphy.

Packing: Each vial contains 1 ml with approximately 0.2mg/ml monoclonal antibody and 1 mg BSA in 20 mM TRIS and 150 mM NaCl, pH 8.0.

Preservative: Merthiolate (0.001%).

Storage: Monoclonal antibodies should be stored below -18Deg C.Repeated thawing and freezing may reduce the antibody activity and should be avoided. Do not use this product beyond the stated expiration date.

Presentation: 100ug in 0.1ml (1mg/ml) 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 0.5   mg/ml Gentamicin Sulfate (as a preservative).

purification: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Purity was >95% Immunoglobulin by SDS-PAGE and contains less than 1% Bovine Immunoglobulin.

Isotype: Mouse IgG1

Immunogen: Human WM-1 (Waldenström's macroglobulinemia) cell line

INFORMATION: Human CD70 is not present on resting lymphocytes. CD70 is expressed after activation on about 20% of T cells and on about 70% of B cells. CD70 is a ligand for CD27 and it is also a member of the tumor necrosis factor gene family. Antibody BU69 recognizes CD70 molecule of approximately 30 kd. Antibody BU69 inhibits T cell proliferation induced by dendritic cells.

STORAGE: Store at 2 - 5 oC. Freeze/Thawing is not   recommended.

PERFORMANCE: 500K cultured Raji human tumor cells were washed and incubated 45 minutes on ice with 80ul of anti-CD70 at a concentration of 20ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC , after which they were washed three times, fixed and analyzed using a BD FACstar plus. Cells stained positive with a mean shift of 0.88 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.

SPECIFICITY The antibody reacts with the 29 kDa moiety which is a type II transmembranous molecule of the TNF family. The antigen is expressed on a large proportion of activated B cells, a few activated T cells but not on resting lymphocytes, platelets or red cells.

PRESENTATION The monoclonal is presented at a concentration of 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.
