SPECIFICITY This antibody reacts with a single chain glycoprotein of 75 kDa. Its main cellular reactivity is with monocytes (upregulated by interferon gamma). Other reactive cells in flow cytometry include interferon gamma stimulated granulocytes, myelomonocytic leukaemias, and myeloid cell lines. Some histiocytes are recognized by this antibody in immunohistochemistry.
APPLICATIONS * Blocks immunoglobulin binding to FcR1 receptor (clone 10.1 is the only FcR1 antibody with this reactivity)* May block antibody dependent cell killing with a potential application in autoimmune disease* Used in the production of bispecific antibodies, and as an indicator of inflammation (FcR1 induced on macrophages by interferon gamma)

PRESENTATION The monoclonal is presented at a concentration of 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.
STORAGE For use within 1 month of purchase store at +4oC, for long term storage aliquot antibody into small volumes and store at -20oC.

PRESENTATION The conjugate is supplied as a 100 test vial in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. For flow cytometry we recommend using 10ml of conjugate to label 106 cells.

Immunogen: Human rheumatoid synovial fluid cells/monocytes
PRODUCTION: Antibody was Protein A purified from (low FBS containing) tissue
culture supernatant. Immunoglobulin was enzymaticly cleaved using Immobilized
Ficin. F(ab')2 was separated from intact antibody and Fc fragments by protein A
and Size exclusion chromatography. Purity was >90% F(ab')2 by SDS-PAGE.
Activity: Antibody 10.1 recognizes the CD64 molecule of 72 kd from gene Fc gRIA. Antibody 10.1 blocks binding of FcgRI to immunoglobin opsonized cells.
STORAGE : Store at 2 - 5oC. Open under aseptic conditions. Freeze/Thawing is not recommended.
INFORMATION: Human CD64 is a high affinity receptor for monomeric human IgG1 and IgG3 which is expressed on macrophages, monocytes, and gamma interferon induced neutrophils. CD64 plays an important role in clearance of immune complexes and in antibody dependent cytoxicity.

Performance: Five x 10 5 cultured U-937 cells were washed and incubated 45 minutes on ice with 80 ml of anti-CD64 F(ab')2 at a concentration of 10 ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse/FITC, after which they were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive (67% net) with a mean shift of 0.69 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.
