SPECIFICITY: The CD56 antogen is a 175-185kDa isoform of the neural cell adhesion molecule (NCAM). It is expressed by lymphocytes mediating non-MHC restricted cytotoxicity and is present on some neural tissues and tumors.

APPLICATIONS: * Recognition of a membrane antigen present on retinoblastoma and other neuroectodermal tumours and tissues.

                             * Immunohistological characterisation of neuroectodermal tumours and in detailed biochemical         investigation of expression of different isoforms of NCAM’s on different tumours.

-This isoform works on frozen sections but NOT in flow cytometry

-Identification of NK cells

-immunoprecipitation studies of CD56 antigen

-Also reacts with porcine antigens

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration of not less than 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml of bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -200C. After thawing they should then be used as a stock solution for up to 30 days. Dilute working solutions are best prepared on the day of use.

SPECIFICITY   CD56 is an isoform of the NCAM molecule that is expressed by all lymphocytes that mediate non-MHC restricted cytotoxicity. MEM 188 is specific for the extracellular domain of NCAM that consists of five C2 set lgSF domains and 2 fibronectin III domains

Antibodies to N-CAM (CD56)

Expression of neural cell adhesion molecules (N-CAM) provides neurons with a means of attaching to and interacting with other cells and the extracellular matrix. Alternative splicing of N-CAM mRNA results in several N-CAM isoforms. The three major isoforms are referred to as 180 kD, 140 kD, and 120 kD. Individual isoforms undergo various post-translational modifications, including the addition of polysialic acid residues to embryonic forms. The highly polysialylated forms of N-CAM present in embryos and neonates are lost during maturation and replaced by less-polysialic  acid-laden mature forms of N-CAM. The functional significance of  alternative forms of N-CAM remain to be fully elucidated. RDI offers N-CAM antibodies recognizing the major isoforms of adult and embryonic N-CAM (N-CAM 13), and embryonic N-CAM (12F8).

-N-CAM 13 recognized the three major isoforms of N-CAM, referred to as 120 kD, 140 kD, and 180 kD. It recognized an extracellular epitope. N-CAM 13 has been shown to react with mouse and rat. Clone 12F11 recognizes the 140 and 180 kD isoforms of N-CAM, but not the 120 kD form. 12F11 recognizes an intra-cellular epitope. 12F11 has been shown to react with human,rat,mouse, and chick  neuronal N-CAM. Applications include western blot analysis, and  immunohistochemistry of frozen tissue sections for light and electron microscopy, and of tissue-cultured neurons. Rat anti-Embryonic N-CAM

Clone 12F8 reacts with the polysialic acid on N-CAM. The epitope recognized is a neuraminidase-sensitive carbohydrate. Additionally, it may cross-react with carbohydrate epitopes that may be expressed in the embryonic than in the adult brain. As such, 12F8 is useful for distinguishing embryonic forms of N-CAM from adult forms which have decreased polysialic acid expression. 12F8 will also react with some non-neuronal cells expressing polysialylated N-CAM, however, it does not appear to recognize mouse NK cells. 12F8 recognized all species tested, from fish through human. Published references include human, rat, mouse, and avian. Applications include western blot analysis, and immunohistochemistry of  frozen tissue sections.
