PURIFICATION METHOD: Protein A affinity chromatography.
SPECIFICITY: CD50 (Icam-3) is the third counter receptor for LFA-1 and is a focal point for leucocyte interactions. The antigen molecule is a N-glycosylated monomer (110kDa) containing intra- chain disulphide bridges which is capable of transducing signals across the plasma membrane. T cell activation with mitogns, anti-CD3 antibodies or PMA results in the phosphorylation of the 5-cytoplasmic serine residues of ICAM-3.

* Studies of cell signal transduction, in particular the relationship between the tyrosine kinases p56lck and p59fyn and ICAM-3
-Studies of the soluble serum form of Human CD50 particularly in systemic lupus erythermatosus (SLE)
-Flow studies of CD50 distribution which is widely found on leucocytes.

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration of not less than 200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml of bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.
STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -20C. After thawing they should then be used as a stock solution for up to 60 days. Dilute working solutions are best prepared on the day of use.

Specificity: This antibody specifically recognizes human ICAM-3 and does
not cross with ICAM-1, ICAM-2 or LFA-1. Not tested for other species.
Neutralization: significantly inhibits binding between LFA-1 and ICAM-3.
ICAM3 is a heavily glycosylated transmembrane 124kDa-molecule with 5 extra-
cellular domains. In contrast to ICAM-1 and ICAM-2 it is particularly expressed on
resting lymphocytes, monocytes, and neutrophils. Icam-3 is not found on
endothelial cells.
Together with ICAM-1 and ICAM-2, ICAM-3 acts as a ligand for LFA-1 and binds this B2-
integrin through its 2 N-terminal domains. All three Icams-molecules own distinct
features during the interaction between leukocytes and their different targets. In
this context ICAM-3 is potentially the most important ligand for LFA-1 in the initiation
of the immune response, e.g. the adhesion of resting T-lymphocytes to antigen presenting
B-cells. After immune cell activation the importance of ICAM-3 decreases because ICAM-1
is upregulated to a much greater extent.
An involvement of ICAM-3 in the T-cell mediated lysis of infected cells and the
allogenetic as well as autologous mixed lymphocyte reaction is discussed.
Application: Anti-ICAM-3 can be used to stain acetone fixed cryostat sections or cell smears. The initial dilution is 1:40 to 1:100 (optimal dilution should be determined by the individual lab for each application. -The antibody may also be used in immunofluorescence. -neutralization: significantly inhibits binding between LFA-1 and ICAM-3.
