purification: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Purity was >95% Immunoglobulin by SDS-PAGE and contains less than 1% Bovine Immunoglobulin.

Isotype: Mouse IgG2a

Immunogen: Stimulated human leukocytes

Specificity: Antibody 158.4D3 recognizes all CD45 molecules containing the A region exon (1,2). Human CD45RA is expressed on all cells of hematopoietic origin, except erythrocytes. CD45RA is a transmembrane tyrosine phosphate which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. The CD45RA isoform predominates on naive/resting T cells and medullary thymocytes.

STORAGE: Store at 2 - 5 oC. Freeze/Thawing is not recommended.

PERFORMANCE: 500K cultured human Raji cells were incubated 45 minutes on ice with 80ul of anti-CD45RA antibody at 5ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC after which they were washed three times, fixed and continued analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 1.86 log10 fluorescent units when compared to a Mouse IgG2a negative control (cat#RDI-MIGG2aX2 $188.00/100ug) at a similar concentration.

PURIFICATION METHOD: Ion exchange chromatography following ammonium sulphate precipitation.


SPECIFICITY: 220 kD MW component of the Leucocyte Common Antigen complex located on some CD4+ T lymphocytes, B lymphocytes and CD8+ lymphocytes. The CD45R+/CD4+ lymphocytes functionally exert suppressor/inducer activity in in vitro assay systems.

Antigen distribution: B-cells (Ig+) > 95%

T-cells (E+) 70 ± 10%

Peripheral blood lymphocytes 68 ± 8%

T-cells (CD4+) 50 ± 10%

Monocytes 50 ± 20%

Thymocytes < 5%

Granulocytes < 1%

* Identification of CD4+ suppressor inducer cells, by direct immunoflourescence staining

* Identification of CD45R+ lymphocytes in cryostat sections of frozen tissue.

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration of 200µg/ ml in phosphate buffered saline containing 10 mM sodium azide and 1 mg/ml of bovine serum albumin. We recommend that each laboratory determine an optimum working titre for use in its particular application.

STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in aloss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -20C. After thawing they should then be used as a stock solution for up to 60 days. Dilute working solutions are best prepared on the day of use.
