INFORMATION: Human CD43 is a heavily O-glycosylated and sialylated transmembrane
protein that is mainly found on T cells, but not on B cells. Antibody DFT1
recognizes epitope A of the CD43 molecule on pMN of about 135 kd (1,3). Antibody
DFT1 partially induces apoptosis in hemopoetic progenitor cells and also
induces homopoetic aggregation.

PERFORMANCE: 500K cultured Jurkat cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD43/FITC at a 1:50 dilution factor. Cells were washed three times, fixed and analyzed on a BD FACS Calibur. Cells stained positive with a mean shift of 1.71 log10 fluorescent units when compared to a Mouse IgG1/FITC negative control (Catalog#RDI-MIGG1FTX2)at a similar concentration. Binding was 97% blocked when cells were pre incubated 10 minutes with 20ul of 0.5 mg/ml anti-CD43 antibody

PURIFICATION METHOD: Protein A affinity chromatography
SPECIFICITY: CD43, the glycoprotein leukosialin (95-115kDa MW ) antigen present
on all T lymphocytes , most B-lymphocytes and on granulocytes, monocytes
and platelets in peripheral blood. The antigen is also expressed cytoplasmically
by macrophages.
The antigen survives formalin fixation and conventional processing to prepare
wax sections. Staining may be achieved either with or without mild proteolysis
of wax sections prior to staining.
APPLICATIONS:
* Identification of normal and malignant T lymphocytes in cryostat sections of frozen tissues.
-Identification of CD43 positive cells on formalin fixed paraffin wax embedded tissue sections.
* Leukaemia and lymphoma phenotyping
* Studies of the interaction between ICAM-1 (CD54) and the CD43 extracellular domain.

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration
of not less than 200µg/2ml in phosphate buffered saline containing 10mM
sodium azide and 1mg/ml of bovine serum albumin. We recommend that each
laboratory determine an optimum working titre for use in its particular
application.
STORAGE: Purified monoclonal antibodies are generally stable for long periods
at 4oC. However repeated warming to room temperature and re-cooling may result
in a loss of activity and hence effective working titre. It is recommended
that monoclonal antibodies are aliquotted upon receipt and then stored at
-20C. After thawing they should then be used as a stock solution for up to
60 days. Dilute working solutions are best prepared on the day of use.
