PURIFICATION METHOD: Ion exchange chromatography
SPECIFICITY: The antibody reacts with CD40, a 48kDa glycoprotein which is found on all mature peripheral B lymphocytes but is absent from terminally differentiated plasma cells.. It is present on some epithelial cells, carcinomas and lymphoid dendritic cells.
APPLICATIONS:
* Quantitative determination of peripheral B cells in blood
* Identification of CD40 positive cells in tissue sections including paraffin embedded sections.
* The antibody promotes homotypic adhesion and gives a strong cycle progression signal
* The CD40 antigen is a possible receptor for a soluble product/cellular ligand supporting proliferation of B cells

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration of 200ug in 2ml phosphate buffered saline solution with 1mg/ml BSA and 10mM NaN3 .We recommend that each laboratory determine an optimum working titre for use in its particular application.
STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -200C. After thawing they should then be used as a stock solution for up to 60 days. Dilute working solutions are best prepared on the day of use.

Immunogen: A 20 residue peptide corresponding to amino acids 258-277 of the human CD40 sequence (CQPVTQEDGKESRISVQERQ) coupled to KLH.
Use: Working dilution: 1:250 for immunoblotting (colorimetric) > with chemiluminescence. Titer for each application.
Specificity: Recognizes a 43 kDa band corresponding to the expected molecular weight of CD40 by SDS-PAGE. Reacts with CD40 in human, mouse, rat, pig, and monkey cells.
Storage: -20°C. Recommend aliquot and store at -20 DEG C. Avoid frequent freeze thaw cycles. At first thaw, tighten cap and centrifuge for 1 minute 1000rpm to concnetrate material in vial.

Activity: Reacts with human CD40L. This antibody immunoprecipitates a CD40L
(gp39) molecule of 36 kd. The antibody will stain
activated CD3+human PBL and will functionally block MLR, sgp30 induced B cell
proliferation and T cell dependent B cell differentiation.
Application: -Indirect fluorescence (approx 0.8ug/500K cells) -Briefly, ficoll
prepared human peripheral white blood cells were stimulated by incubating 6
hours at 5 million cells/ml in RPMI 10% FBS media including 3ug/ml
phytohemagglutinin-P and 1ng/ml Phorbol 12- Myristate 13-acetate. 500K cells
were then washed and incubated 45 minutes on ice with 80ul of product at
10ug/ml. Cells were washed 2X and incubated with 50ul of secondary antibody and
then washed 3X. A net of 5% of population of the cells stained positive with a
mean shift of 306 fluorescent units compared to the IgG1 negative control.
-Blocking experiments-MLR (mixed Lymphocyte Response) can be blocked using approx 10ug/ml when 300K human PBL's(responders) are cultured with 100K irradiated allogenic human PBL's (stimulators). Inhibition can be observed by day 5 when using a 3H thymidine pulse to monitor proliferation. B cell differentiation induced by activated T cells and Il-2 can be measured by Ig production of IgD+ naive B cells.
Monoclonal anti-CD40L significantly inhibits this Ig production at
levels of 1-10ug/ml.

Activity: Reacts with CD40-Ligand (newly clustered as CD154 in VIth workshop), a 39 kd type II membrane glycoprotein expressed on activated T cells. CD40-CD40L interaction plays a very important role in T cell depenednet B cell proliferation, differenetiation, and memory cell formation. CD40L MAB partially blocks T cell-B cell interaction affecting subsequent proliferation, IL-2R expression and differentiation on B cells. In addition, blocking of CD40-CD40L, interaction has been demonstrated with soluble CD40, resulting in the inhibition of immunoglobulin isotype switching. It has been reported that patients with X linked hyper-IgM syndrome have defective expression of functional CD40L due to a defective gene that encodes CD40L.
