Reactivity: The monoclonal antibody is directed against the CD4-antigen
(T4-antigen),which is expressed on human peripheral T
lymphocytes and 80% of thymocytes. The monoclonal antibody reacts on a low
level with human monocytes and macrophages. The monoclonal antibody does not
react with B-cells ,granulocytes and thrombocytes.
The monoclonal antibody binds to the T lymphocytes with helper cell activity in pokeweed mitogen stimulated immunoglobulin production, as was shown in separation experiments (i.e."panning").
Applications: Monitoring of T-cell subset numbers in peripheral blood of patients with immunological diseases.
Analysis of immuno-regulatory T-cell subsets involved in helper/inducer functions.
Characterization of leukaemias and lymphomas.
Identification of T-cells in tissue.
Methods: Indirect immunofluorescence staining with analysis by FACS or
fluorescence microscopy.

This clone has been derived from hybridization of SP2/0 cells with spleen cells of a (BALB.x x A/J) mouse immunized with human T-lymphocytes.
Mouse isotype:The monoclonal antibody is of the mouse subclass IgG2a.
Source: Ascites fluid of tumour bearing BALB/c mice.
Purification: Ammoniumsulphate precipitation and ion exchange chromatography.
Packing: Each vial contains 1 ml with approximately 0.1mg/ml monoclonal
antibody and 10 mg BSA in 20mM TRIS and 150 mM NaCI, pH 8.0.
Preservative: Merthiolate (0.001%).
Storage: Monoclonal antibodies should be stored below -18C. Repeated thawing and freezing may reduce the antibody activity and should be avoided.

PURIFICATION METHOD: Ion exchange chromatography
SPECIFICITY: 60Kd MW lymphocyte surface antigen identified by monoclonal
antibodies belonging to the CD4 cluster and present on 54% of peripheral blood
T lymphocytes, 50% of thymocytes and some malignant cells of T cell origin.
Normal B lymphocytes, monocytes or granulocytes do not express surface CD4
antigen although cytoplasmic expression has been observed in
monocytes/macrophages.
The CD4 positive T lymphocyte subpopulation has been characterised functionally as comprising helper cells active in amplification of immune responses.

T-cells (E +) 68 ± 8%
B-cells (Ig +) < 1%

APPLICATIONS:
* Identification of CD4 positive T helper/inducer lymphocytes by indirect immunofluorescence staining.

* Isolation or removal of helper/inducer T lymphocytes by FACS or panning techniques

PRESENTATION: Each monoclonal is delivered in a sealed vial at a concentration
of not less than 200µg/2ml in phosphate buffered saline containing 10mM
sodium azide and 1mg/ml of bovine serum albumin. We recommend that each
laboratory determine an optimum working titre for use in its particular
application.
STORAGE: Purified monoclonal antibodies are generally stable for long periods
at 4oC. However repeated warming to room temperature and re-cooling may result
in a loss of activity and hence effective working titre. It is recommended that
monoclonal antibodies are aliquotted upon receipt and then stored at -20C.
After thawing they should then be used as a stock solution for up to 60 days.
Dilute working solutions are best prepared on the day of use.
