PRODUCTION: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Purity was >95% Immunoglobulin by SDS-PAGE and contains less than 1% Bovine Immunoglobulin.
INFORMATION: Human CD21 is a receptor for complement fragments C3d, C3dg or ic3D and also EBV . CD21 is also a ligand for CD23 and plays a role in IgE synthesis. Antibody BU33 recognizes the CD21 molecule of about 145 kd.Antibody BU33 inhibits binding to CD23.

STORAGE: Store at 2 - 5oC. Freeze/Thawing is not recommended.
PERFORMANCE: 500K cultured HPB-MLT cells were washed and incubated 45 minutes on ice with 80 ul of anti-human CD21 (CR2) at 20 ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC , after which they were washed three times, fixed and analyzed continued on back side using a BD FACstar plus. Cells stained positive with a mean shift of 1.17 log10 fluorescent units when compared to a Mouse IgG1 negative control (Catalog #RDI-MIGG1X2, $188.00/0.1mg) at a similar concentration.

This antibody has been clustered toCD21 in the fourth international Workshop
on Human White Cell differentiation Antigens in Vienna.
Isotype Mouse, IgG2a.
Source Mouse ascites fluid.
Purification Ammoniumsulphate precipitation followed by ion exchange
chromatography.
Conjugation Conjugated with fluorescein iso thiocyanate isomer 1 (FITC).
Molecular F/P ratio between 5.5 - 9.5.
Packing Each vial contains 1 ml FITC conjugated monoclonal antibody 2 mg
BSA in PBS.
Preservative 0.1% NaN3.
Storage and stability Monoclonal antibodies should be stored in the dark
at 2-8°C. The reagent is stable until the expiry date stated on the
vial label.
Major reactivity The monoclonal antibody is directed against the CD21-antigen,
which is expressed on normal Ig-positive B-cells from peripheral blood and
lymphoid tissues and on dendritic cells of germinal centres. Its expression
is lost on activated B-cells. The distinct distribution among B-cell malignancies
differs from other B-cell markers e.g. CD19 and CD20.
The monoclonal antibody reacts positive with some cases of childhood T-cell
acute lymphoblastic leukaemia (T-ALL).
The monoclonal antibody does not react with mature T- and NK-cells and non lymphoid haemopoietic cells.
Molecular mass 140-145 kDa.
Application Identification of CD21 positive lymphocytes cells.
Identification of normal and malignant CD21 positive B-lymphocytes on frozen sections.
Methods Direct immunofluorescence staining with analysis by flowcytometry or fluorescence microscopy.
