Activity: Reacts with human CD166 (Alcam, activated leucocyte cell adhesion
molecule. mr 100,000 with five Ig-like domains), a type 1 glycoprotein expressed
on activated T cells, B cells and monocytes and appears to be the ligand
for CD6. Human CD166 may be important for activation of T cells. Alcam is
a ligand for CD6 and Mab from clone 3A6 blocks binding of CD6 to CD166.

bodies, fibroblasts, endothelial cells, epidermal keratinocytes and neurons.

Application: 500K cultured Raji cells per tube were washed and preincubated
5 minutes with 20ul of 250ug/ml human IgG(to block non specific binding)
after which they were incubated 45 minutes on ice with 80ul of the anti-CD166
antibody at a concentration of 5ug/ml. Cells were washed twice and incubated
with 50ul of secondary reagent Goat anti-mouse IgG:FITC, after which they
were washed three times, fixed and analyzed using a BD FACSCalibur. Cells
stained positive with a mean shift of 0.87 log 10 fluorescent units when
compared to a mouse IgG1 negative control at a similar concentration.

on ice with 80ul of the anti-CD166:Fitc conjugated diluted 1:50. Cells were
washed three times, fixed and analyzed by Facs. Cells stained positive with
a mean shift of 0.71log10 fluorescent units when compared to the mouse IgG1:FITC
negative control. Binding was blocked 98% when cells were pre-incubated 10
minutes with 20ul of 0.5mg/ml anti-CD166 purified antibody.
