Activity: Reacts with 4-1BB, a 30kDa glycoprotein expressed on activated
T cells. 4-1BB can be detected on Con A or PHA- stimulated peripheral blood
cells (CD4+ and CD8+) and on CEM cell (human T-cell leukemia cell line) following
2 day PMA and ionomycin stimulation, but not on resting T cells. 4-1BB may
be a participant in T-cell activation. It has been reported to be responsible
for rescuing T cells from activation-induced apoptosis, upregulation of TH-1
type, down regulation of TH-2 type cytokine responses, and induction of cell
adhesion to fibronectin.

Background: Human CD137 (4-1BB) is expressed on activated T cells within
24-48 hours of activation. CD137 is a type I membrane protein and a member
of the tumor necrosis factor receptor superfamily. CD137 appears to be important
for T cell proliferation and survival and induces monocyte activation.

Activity: This human CD137 Ig fusion protein blocks binding of an anti-human
CD137 To activated CEM human tumor cells and also binds to CD137 Ligand on
Raji cells.

Assay: 500K human Raji cells were washed and incubated 45 minutes on ice with 80ul of CD137-huIg at 5ug/ml. Cells were washed twice and incubated with 50ul of secondary antibody (Goat anti-human IgG:FITC) after which they were washed three times, fixed and analyzed by Facs. Cells stained positive with a mean shift of 0.48 log10 fluorescent units when compared to a buffer control at a similar concentration. Binding was blocked when reagent was preincubated 30 minutes with CD137L-muCD8 at a concentration of 50ug/ml.
