Immunogen: synthetic peptide derived from aa 416-430 of human tubulin beta
Species Reactivity: Human, Cow, Pig, Rat,Mouse, Guinea pig, Gerbil, Frog, Chicken, Yeast, and Fungi. sea urchin, myxamoeba and plasmodium.Others-not tested.
Localization: Cytoplasmic
Use: -western blot (1ug/ml for 2 hours RT)
-histochemistry: frozen sections and formalin fixed paraffin sections (no special pretreatment required) 1-2ug/ml
Positive Control: Skin or lung
Specificity: Recognizes a protein of 55kDa, identified as tubulin Beta. Its epitope localizes between aa 416-430. although not tested for this activity, a similar Monoclonal ab clone DM1B (no longer available) did not cause the 10-nm filaments to collapse into large lateral aggregates collecting in the cell periphery or tight juxtanuclear caps. It does not block microtubule assembly. It does not inhibit polymerization or depolymerization of platelet tubulin in vitro. It blocks (by 70-80%) the ability of tubulin dimers (with GppNHp bound) to promote a stable inhibition of adenylyl cyclase.
