Immunogen Laminin isolated from Engelbrecht-Holm-Swarm (EHS)-mouse sarcoma
Introduction Laminin is a glycoprotein (Mr 850 - 1.000 kDa, consisting of 3 glycosylated polypeptide chains with molecular weights of 440 and 225 (2x) kDa) produced by various human epithelial and mesenchymal cells, and forms an extracellular matrix of thin filaments. In normal tissues, laminin is invariably present in all basal laminas surrounding muscle, nerve, fat and decidua cells and separates epithelial and endothelial cells from abutting connective tissues. Laminin has also been identified within the cytoplasm of breast epithelia, stromal cells of the endometrium, and within endothelial, bile duct epithelial and mesenchymal cells of the liver. Laminin has been found to be involved in cellular activities such as adhesion, spreading, differentiation, polarization, proliferation, locomotion, tissue invasion and chemotactic responses.
Clinical Application/ Specificity The antibody reacts with a 200 kDa protein band in immunoblotting. No cross reaction was obtained with human type I, III, IV and V collagen in immunohistochemstry.

Mouse Laminin Antigen

Presentation: 2mg in 2.1ml of 0.05M Tris-HCl, pH 7.4, 0.15M NaCl. Frozen solution.
Source: Purified from basement membrane of the Engelbreth-Holm-Swarm (EHS) mouse tumor using salt precipitation and ion-exchange chromotography.
Use: Thaw frozen product at 4 C and keep on ice thereafter. Dilute with sterile buffer or serum-free medium to approximately 20 g/ml. Add directly to culture vessels to achieve 2.5-10 g/cm 2 . Allow the matrix proteins to adsorb to the vessel for one hour at 37 C or overnight at 4 C prior to removing and adding medium.
Sterility: This lot of laminin was tested for the presence of bacteria, fungi and mycoplasma.
Storage: Stable for 1 year at -70 C from date of shipment. Aliquot to avoid repeated freezing and thawing by dispensing into appropriate aliquots if the material is not to be used all at once. Dilute in any physiological buffer or culture medium.

Quality Control Testing
Attachment Assay: This lot of laminin was tested for the ability to promote cell attachment using PC12 rat pheochromocytoma cells in an attachment assay. The optimal range was found to be 2.5-10 g/cm 2 . Laminin Attachment Assay Protocol
1. Coat 6 well dishes with laminin, at a concentration of 2.5-10 g/cm
2 . Incubate 1 hour at 37 C or overnight at 4 C. Aspirate and rinse 2 times with PBS before adding cell suspension.
3. Prepare PC-12 cell suspension at 2x10 4 cells/ml in RPMI-1640/10% horse serum/5% FBS. Add 2ml of cell suspension to each well.
4. Prepare NGF as described on certificate of analysis to a final concentration of 20ng/ml. 5. Incubate plates at 37 C for 3-5 days.
6. Record neurite outgrowth at each laminin concentration.
