Mouse anti-PMCA ATPase Monoclonal ab

Background: The Ca2+ pump of the plasma membrane, termed PMCA ATPase pumps
CA2+ from the cytosol to the extracellular space. This membrane bound enzyme
is related to a number of other ATPases including the SERCA ATPase and the
NA K Pump. Studies indicate there are four known isoforms of the PMCA ATPase
which are encoded by a multigene family.

package: 100ul of diluted ascities fluid with 0.05% sodium azide added
antigen: produced from a fusion of FO myeloma cells and spleen cells from
Balb/c mice immunized with purified human erythrocyte CA2+ ATPase.
Specificity: epitope mapping between amino acids 724-783 of the human erythrocyte
CA2+ pump. This epitope is located in the highly conserved hinge region which
is on the intracellular loop of PMCA ATPase between the putative transmembrane
domains M4 and M5. This clone reacts with all four known isoforms of PMCA
ATPase and a wide range of species and tissues as shown below.
Human: erythrocyte, kidney, placenta, choroid plexus
Primate: kidney
Canine: kidney
Feline: erythrocyte, choroid plexus
Chicken: oviduct, intestine
Rat: erythrocyte, liver, kidney, aortic
endothelial, intestine, placenta,
enamel/dentin, brain, choroid plexus.
Hamster: erythrocyte, liver, kidney, aortic
endothelial, intestine, brain
Eel: kidney
RABBIT: kidney
Use:
-Western blot 1:1000 recognizes approx 140kDa band representing PMCA ATPase and bands at 95Kda and 180Kda which are probably products of aggregation and/or natural proteolytic products of the pump.
-Immunohistochemistry: 1:500 staining depends upon tissue being studied and location of the pump. Suitable for both frozen and paraffin embedded tissues.
-Immunoprecipitation
Storage: Store at -20 DEG C upon receipt. Avoid frequent freeze thaw cycles.
Recommend aliquoting. BEFORE OPENING, suggest centrifuging for 1 minute at
1000 RPM to concentrate material in vial.
