The hybridoma from which this antibody was produced was the product of a fusion between Sp2/0 myeloma cells and splenocytes from a BALB/c mouse immunized with a synthetic peptide whose sequence was derived from the ER-DNA binding, amino acids 247-261. The DNA-binding domain of the ER spans amino acids 185-261.

Isotype: mIgG1
Specificity: This clone reacts with the estrogen receptor from human, calf,
rat and mouse tissues and does not cross with the progesterone receptor or
glucocorticoid receptor.
-reacts with both the steroid occupied and unoccupied forms of ER and does
not bind to native, steroid binding from of ER. Some studies have shown that
this clone does not inhibit binding of the ER to DNA and in fact the antibody
is released from the ER upon DNA binding of the receptor.
USE: immunolocalization (5ug/ml immunoprecipitation (must titer)
Optimal dilutions must be obtained for all applications.
Storage: -20 C. Avoid repeated freeze/thaw cycles.

Immunogen: produced by immunizing New Zealand white rabbits with a synthetic
peptide corresponding to the N- terminal amino acid residues 55-70 of
rat and mouse ERB conjugated to KLH. This sequence is completely conserved
in human and marmoset ERB protein.

Background: Steroid receptors are ligand-dependent intracellular proteins
that stimulate transcription of specific genes by binding to specific DNA
sequences following activation by the appropriate hormone. A receptor for
estrogen (ER) was first cloned in 1987 and has been shown to be expressed
and transcriptionally active in a number of tissues including ovary, testis,
prostate and brain. Until recentlty, only the "classical" ER (ERa) has been
known. However, a new estrogen binding receptor,referred to as ERB, has recently
been cloned in rat and humans.
ERB has been shown to be a high affinity estrogen binding protein
capable of initiating transcription of genes under the control of estrogen
response elements (ERE;s). RNA distribution studies have shown ERB is transcribed
in human thymus, spleen, ovary and testis as well as rat ovary and prostate.
This distribution overlaps with but is not identical to the tissue distribution
of ERa RNA. While ERB RNA distribution has been studied, little is known
about protein distribution under various hormone conditions. ERB protein
distribution may differ from RNA distribution and from ERa protein given
the complex and differing activation, dimerization and translocation pathways
exhibited by various steroid receptors under various ligand states.
Reactivity: Reacts with various cell types in normal and ERa knock- out mice
and normal rats. No immunostaining is observed with pre-immune serum and
specific staining is eliminated when the antibody is pre-incubated with the
immunizing peptide. No immuno-cross-reactivity has been observed with ERa
expressed in baculovirus.
Uses: -immunohistochemistry and gel super-shift procedures to detect ERB.
Produces cytoplasmic staining in Purkinje neurons of rat cerebellum, cells
in the murine pituitary gland, and cynomolgus monkey cortical amygdala. In
addition ERB immunostaining has been observed in mouse Leydig cells and in
the transitional epithelium of the urinary bladder in male and female wild-type
and ERa knock-out mice. This antibody efficiently supershifts rat ERB/ERE
(vitellogenin xenopus A2 gene). This antibody does not detect ERB protein
in Western blotting applications.
Dilution: Various assay conditions require that the optimal wroking
concentrations be determined by serial dilution of this product. Suggested
starting dilution is:
