COLLAGEN ANTIBODIES
(anti-Human and others as indicated)
RDI Division of Fitzgerald Industries Intl offers a wide line of antibodies. Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Please inquire for other applications or types of antibodies not listed below.

Background: Recognizes collagen type II and shows no reaction with types I, or III. In western blot, this clone reacts with the TC(a) fragment of lathyritic type II collagen after digestion with mammalian collagenase. It also reacts with pepsin digested type II collagen. The epitope for clone 6B3 is localized in the triple helix of type II collagen. Immunoblotting of CnBr peptides of collagen II shows that clone 6B3 Mab reacts with CB 11 fragment which is the site of immunohgenic and arthritogenic epitopes alon the intact II molecule. In western blot (but not in Elisa) this Mab recognizes a1(II) as well as a3(Xi) chain both of which have identical primary structure.

Background: Recognizes collagen type II and shows no reaction with types I, III, IV, V, VI, IX, X or XI. In western blot, this clone reacts with the TC(a) fragment of lathyritic ype II collagen after digestion with mammalian collagenase. It does not react with pepsin-digested tyep II collagen. The epitope for this Mab is localized to the aminotelopeptide of the type II collagen. Precise mapping of clone 5B2.5 is reported to be GGFDEK, slightly preceding the lysine residue involved in crosslinking. Two germinal residues constitue a key structural element of the epitope. The epitope for this Mab is lost after digestion of lathyritic type II collagen with recombinant stromelysin.

Reactivity: human (others not tested)
Uses: Immunohistology (Formalin/paraffin) (Use Ab at 1:50-1:100 for 30 min at
RT) (Staining of formalin/paraffin tissues is enhanced by digestion of tissue
sections with Protease XXV at 1mg/ml PBS for 5 minutes at 37°C
Positive Control: Skin.
Localization: Basement membrane
Background: Collagen IV is a major constituent of the basement membranes along
with laminins and enactins. It is composed of á 1(IV) chain and á 2(IV) chain
in 2:1 ratio . It can form insoluble fibers with high tensile strength.
Antibody to collagen IV is useful in detecting the loss of parts of basement
membrane in carcinomas. This MAB cocktail is highly specific to type IV
collagen. In kidney, it reacts with glomerular and tubular basement membranes,
parts of mesangial matrix and the Bowman’s capsule. It also reacts with basal
lamina of capillaries as well as basement membranes in a variety of tissues.
This Mab cocktail is excellent for the staining of routine formalin-fixed,
paraffin-embedded tissues.

bullous lesions in dermatopathology. No cross reactivity with other collagens
at optimal dilutions. In immunoblotting1, a slight cross reactivity with
collagen type V is observed.

Cellular Localization: Epidermal basement membrane.
Comments: Mab shows no cross-reaction with Type I, II, III, IV, and V collagens, laminin, and fibronectin. It reacts with the basement membrane zone of stratified squamous epithelia. Ab-1 is useful in differentiating invasive from non-invasive melanoma through clear visualization of appearance and integrity of epidermal basement membrane. This Mab is also helpful in the identification of recessive Dystrophic Epidermolysis Bullosa (RDEB) in which Type VII collagen is defective.

Reactivity: Reacts with human, mouse, and rat collagen type X. Exhibits slight
cross-reactivity to fibronectin and collagen types II and IX. Does not cross
react with collagen types I, III, or XI.
Description: Undiluted polyclonal antibody recognizing type X collagen.
Collagens are major fibrous structural elements of cartilage, tendon, bone,
skin, lung, and blood vessels. Type X collagen, a heterotrimer consisting of
three a1(X)subunits, is found in hypertrophic and mineralizing cartilage. Type
X collagen provides a transition to bone by allowing removal of hypertrophic
cartilage and calcification. Type X collagen is believed to be involved in
skeletal growth, maintenance, and repair.
Uses: -Immunoblotting:1:100-1:300 with chemiluminescence detection
-Immunofluorescence:1:20-1:40
-immunohistochemistry:1:30-1:60, frozen sections require enzymatic or 41 DEG C heat treatment to unwind the triple helical structure.
-immunoprecipitation1:20-1:60
Storage: At first thaw, tighten cap , centrifuge 1-2 minutes 1000rpm to
concentrate antibody in vial. Recommend making working aliquots. Avoid frequent
freeze thaw cycles. Additional of 0.1% soidum azide or other preservative
recommneded if suitable for your application.

Staining is further enhanced if heat-induced epitope retrieval is followed by digestion of tissue sections with 0.05% hyaluronidase at 37 0 C for 30 min].

Background: Type X collagen is a non-fibrillar, short-chain, network-forming collagen found in fetal hypertrophic cartilage in the growth zones of long bones, vertebrae, and ribs. It is a short collagen with a triple helical core half the length of the fibril-forming collagen type I, II, and III and a globular domain at the carboxyl end (NC-1 domain) through which the molecules assemble into an hexagonal meshwork. The function of collagen X, a unique homotrimer synthesised by hypertrophicchondrocytes, is not known but its localization and transient expression at sites of calcification suggest that it is likely to be associated with events in the early stages of endochondral bone formation.
