RDI Divison of Fitzgerald Industries Intl  offers a wide line of  antibodies. Since no one antibody works best for all applications (neutralization, blotting, histochemistry, ELISA, etc), we offer many different types of antibodies to help solve this problem. Rabbit anti-Calreticulin ab (human/ rabbit/ rat)

Background: Calreticulin is the major binding protein found in smooth muscle sarcoplasmic reticulum (SR) and non-muscle endoplasmic reticulum (ER) membranes. This protein was originally identified in SR membranes and plays a minor role in calcium storage in skeletal and cardiac muscle SR. Calreticulin is also known as calregulin, CRP55, CaBP3, calsequestrin-like protein and R)/SS-A antigen. Calreticulin binds calcium with low affinity and high capacity, however it also exhibits a single high affinity binding site.

The highly conserved sequence Lys-Asp-Glu-Leu(KDEL) is present at the C-terminus of calreticulin and other resident ER proteins including glucose regulated protein 78 (GRP78), GRP94 and protein disulfide isomerase (PDI). This sequence is responsible for the retention of newly synthesized proteins within the ER lumen. This retention is reported to be mediated by a nuclear hormone receptors and may also act as a molecular chaperone.

antigen: produced by immunizing New Zealand white rabbits with recombinant human calreticulin produced in Baculovirus insect cell system.

Specificity: Detects calreticulin in human, rabbit, rat and xenopus non muscle ER.

Use:

-Western blot:1:1000 starting dilution By western blot, recognizes a band at approx 60 kDa and a faint band at 95 Kda which is also seen in pre-immune serum.

-In immunofluorescence (1:1000), produces a fine granular staining consistent with ER localization (rat brain cortex and rat brain hippocampus).

-immunoprecipitation (1:100)

ALL titers must be optimized experimentally.

Storage: Store at -20 DEG C upon receipt. Avoid frequent freeze thaw cycles. Recommend aliquoting. BEFORE OPENING, suggest centrifuging for 1 minute at 1000 RPM to concentrate material in vial.
