Antigen: activated Lewis rat derived microglial cells
Fusion: spleen cells from immunized Balb/c mice were fused with cells of mouse
SP2 myeloma cell line.
CLONE: TLD-3A12
Activity: Reacts with rat (CD31)Pecam-1. Antibody
gives positive reaction at high dilutions against recombinant rat PECAM-1.
Reactivity with other species not determined. Non reactive
with human CD31. CD31 is an approx 130kda integral membrane
glycoprotein, a member of the immunoglobulin superfamily, that mediates
homophilic and heterophilic cell-cell adhesion. CD31 is expressed on
endothelial cells, platelets, and subsets of leukocytes. In the human and
mouse, multiple alternatively spliced isoforms have been identified and this
alternative splicing may be involved in the regulation of ligand specificity.
CD31 mediated endothelial cell- cell interaction are involved in angiogenesis
in the mouse and rat. In addition, PECAm-1 has been demonstrated to play an
important role in extravasation of leukocytes invivo. Mab from clone TLD-3A12
partially blocks the proliferative response of antigen-specific CD4+ T cells to
antigen presenting cells and relevant antigen. This antibody also cross
reacts with Pig endothelial cells and platelets as determined by histochemistry
on acetone fixed frozen sections and flow cytometry.
Application: -Indirect fluorescence (approx 1ug/million cells)
-histochemistry on acetone fixed frozen sections (approx 5-10ug/ml with ABC/Dab detection) rat spleen as positive control continuned on back side
-for paraffin sections recommend zinc fixed sections
-western blotting
-immunoassay
-immunoprecipitation
-in vitro blocking using azide free, sterile material.
-titers must be determined for each application
