Presentation: Purified antibody fraction from rabbit anti-serum. Prepared in 10mM PBS, pH 7.4, with 0.2% BSA and 0.09% sodium azide.

Epitope: Not determined

Reactivity: Human, Cow, Mouse, and Rat. Others-not known.

Immunogen: Ubiquitin isolated from cow erythrocytes

Uses: -Western blot: 1:100 for 2 hrs RT (approx 8.5kda)

   -for paraffin sections, requires boiling tissue sections in 10mM citrate buffer pH6.0 for 10-20 minutes followed by cooling at RT for 20 minutes. -approx ab dilution 1:50-1:100 for 1 hr RT

Positive Control: Human brain from patient with Alzheimers disease

Localization: Cellular Localization: Nuclear, cytoplasmic, and cell membrane.

Background: Ubiquitin (Ub) plays a very important role in regulated non-lysosomal ATP dependent protein degradation. The protein to be degraded is conjugated to Ub and the ubiquinated protein is then selectively degraded by a 26S complex, multicatalytic cytosolic and nuclear protease termed proteasome. The Ub-proteasome proteolytic pathway, which is a complex process, is implicated to be of great importance for regulating numerous cellular processes.

Background: Ubiquitin protein conjugates associated with filamentous inclusions or in dot like strcutures (lyosome-releated organelles) are characteristic of the major human idiopathic neurodegenerative diseases. Dot-like strcutures are aslo a feature of the transmissable neurodegenerative diseases.This antibody detects neurofibrillart TANGLES in Alzheimer's  disease, Lewy bodies in Lewy Body disease, Rosenthal fibres in astrocytomas, Mallory bodies in Alcoholic Liver Disease. Also detects dot-like structures in the white matter of normal aging brain.

Antigen: ubiquitin conjugated with glutaraldehyde cross linked to keyhole limpet haemocyanin.

Activity: Stains human, cow, mouse and rat ubiquitin. Positive staining can be observed in brain sections from patients with Alzheimer's disease. Stains Cytoplasmic filamentous inclusions and dot-like immunoreactivity.

Application -paraffin wax embedded tissue and frozen sections. approx 1:500 dilution. Incubate 60 minutes at room temp, visualize with standard ABC techniques.
