Since no one antibody works best for all applications (neutralization, blotting, histochemistry, ELISA, etc), we offer many different types of antibodies to help solve this problem. Background: Synapsin (synapsins Ia and Ib, Mw 80,000 and 77,000 are collectively
referred to as synapsin I) is a neuron specific protein that is specifically
localized to nerve terminals. the synapsin antibody is an excellent marker
for synaptic terminals and it can be used to estimate synaptic density and
or synaptogenesis.
Specificity:* recognizes human, rat, and bovine Synapsin I
Presentation: 1 vial lyophilized (10ug affinity purified antibody
from PBS
Host: rabbit
Antigen: synapsin I (Ia & Ib) purified from bovine brain
Reconstitution: add 50ul PBS, allow to set at least 30 minutes, mix well,
centrifuge 1-2 minutes at 1000 RPM to concentrate material in vial.
USES: -western blot:initial dilution 1:2000 with enhanced chemiluminescence
detection. (sees a doublet).
-Immunocytochemistry:1:500-1:2500
-Elisa: 1:2500-1:10K
-immunoprecipitation: 1 microgram will quantitatively immunoprecipitate all synapsin in 200ug rat brain.
Note:Optimize titers for each use
Storage: Store lyophilized material at 4 DEG C. Store reconstituted material
at 4 DEG C for immediate use. For long term use, recommend aliquoting and
store at -20 DEG C. Avoid frequent freeze thaw cycles. Store in non frost
free freezer.

Sample Histochemistry Protocol:
a) 30mM sections of rat brain (fixed with 4% paraformaldehyde in PNS) are washed several times in PBS and blocked in a solution of 1% horse serum and 0.3% Triton-X-100 for 1 hour. The tissue is then incubated overnight at 4 DEG C. Higher dilutions may be used for regions with high synaptic density (as cortex and hippocampus), lower dilutions should be use in regions with low synaptic density (as PNS, spinal cord). Sections are washed in PBS and incubated for 2 hours in preabsorbed secondary antibody.
