A goat antiserum to a synthetic peptide that corresponds to amino acids 31-47
of the N-terminus of the Alzheimer disease presenilin-2 protein is currently
available. The presenilin-2 gene was discovered on chromosome 1 and is
responsible for an early onset form of autosomal dominant Alzheimer's disease.
The amino acid sequence is approximately 67% identical with that of the
presenilin-1 protein. It also is a membrane protein that has regions where
mutations have been identified in family members with Alzheimer's disease.
This peptide antibody corresponds to the extracellular portion of this
protein.
The gene sequence of presenilin-2 does not match any known human gene sequences,
except for that of presenilin-1, and the identity and function of both these
proteins remain speculative. This antiserum, has been shown to be immunoreactive
to the presenilin-2 immunizing peptide by ELISA, but does not show
immunoreactivity with the immunizing peptide used for the presenilin-1 antibody.
Currently, this antibody has been shown to label neurons in both AD and normal
control brains using frozen-fixed sections. This antibody should be a valuable
tool for scientists who will be working to discover the function of this
protein in normal individuals and its role in Alzheimer's disease.
