PURIFICATION METHOD: None.
SPECIFICITY: S-100 can be found in glial cells and in tumours of the nervous system, such as schwannomas, ependydomas, and astrogliomas. The antibody also reacts with melanocytes and malignant melanoma, dendritic reticulum cells, adipocytes and chondrocytes. The antibody recognizes an antigenic determinant shared by both the a- and ß-subunit of bovine and human S-100.
APPLICATIONS:
* Useful as a marker in malignant melanoma.
* Reacts with paraffin wax embedded tissue sections.
PRESENTATION: 1.5ml Tissue culture supernatant, which should be diluted 1:10 to 1:100 depending on fixation and detection procedures to be used. The preparation is supplied containing 0.1% sodium azide as preservative.
STORAGE: Purified monoclonal antibodies are generally stable for long periods at 4oC. However repeated warming to room temperature and re-cooling may result in a loss of activity and hence effective working titre. It is recommended that monoclonal antibodies are aliquotted upon receipt and then stored at -20C. After thawing they should then be used as a stock solution for up to 60 days. Dilute working solutions are best prepared on the day of use.

Background: Recognizes proteins of 21-24kDa, identified as the A & B subunits of S100 protein. S100 belongs to the family of calcium binding proteins such as calmodulin and trop0onin C. S100A is composed of an alpha and beta chain whereas S100B is composed of two beta chains. Antibody to S100 stains Schwnnomas, ependymomas, astrogliomas, almost all benign and malignant melanomas and their metasases. S100 protein is also expressed in the antigen presenting cells such as the Langerhans cells in skin and interdigitating reticulum cells in the paracortex of lymph nodes. The diagnosis of Histocytosis X is confirmed by S100 staining. S100 protein is highly soluble and may be eluted from frozen tissue during staining.
