MUTANT C-TERMINAL PEPTIDE ANTISERUM

-A goat antiserum to a synthetic peptide that corresponds to amino acids
339-348 of the C-terminus of the Alzheimer disease amyloid precursor protein
frameshift mutant (APPFM) is currently available. The amyloid precursor protein
in a large membrane protein whose N-terminus projects into the extracellular
space. In Alzheimer's disease, this protein is broken down and releases the 42
amino acid b-amyloid protein that is found in senile plaques and vessels.
-A frameshift mutant of the amyloid precursor protein has been found that
results from a GA dinucleotide deletion which results in a frameshift mutation
that gives rise to a truncated amyloid precursor protein with a unique
C-terminus.
-The role of the amyloid precursor protein frameshift mutant in Alzheimer’s
disease is currently unknown. These abnormal proteins may accumulate and lead
to cellular distrubances and neuronal degeneration. This antiserum has been
shown to be immunoreactive to the immunizing peptide by ELISA. The antiserum
will immunolabel dystrophic neurites in senile plaques in formalin-fixed,
paraffin-embedded sections from Alzheimer brain. It also shows immunoreactivity
with neurons containing the APP frameshift mutnat This antibody should be
useful to scientists and neuropathologists who study the role of amyloid in
Alzheimer’s disease.
