Anti-MOUSE
CD CLUSTERED ANTIBODIES

RDI Division of Fitzgerald Industries Intl  offers a wide line of  antibodies. Since no one antibody works best for all applications (flow cytometry, neutralization, blotting, histochemistry, ELISA, etc), we offer many different types of antibodies to help solve this problem. Please inquire for other applications or types of antibodies not listed below. All products are for in vitro research use only-not for use in or on humans or animals-not for use in diagnostics. Price/availability/specifications subject to change without notice.

Anti-MOUSE  CD95L (FAS-L)  Antibodies

DATA SHEET:    mouse anti-MOUSE CD95L FAS-L clone K10

Catalog#: RDI-MCD95L-K10    $531.00/vial

also available: PE labeled cat#RDI-MCD95L-K10PE $625.00/200ug

              azide free purified cat#RDI-MCD95L-K10X $750.00/0.5mg

Package Size: -500ug purified in 50mM Tris-Hcl (pH 8.0), 0.15M   NaCl and 0.1% sodium azide.

-200ug conjugated in 10mM PBS pH 7.2 with 0.1% NaN3

(sodium azide is toxic, dilute azide compounds with copious amounts of water when disposing in drain to avoid accumulation of explosive deposits-avoid contact with acids.)

Prep: purified from tissue culture supernatant via affinity chromatography.

Species: mouse IgG2b (mouse type C57BL/6 gld/gld)

CLONE: K10 also referenced as KAY-10

Activity: Reacts with Fas-Ligand expressed on activated T lymphocytes of selected strains of mice (C57BL/6, C3H, MRL, NOD, NZB, NZW and SJL). It does not react with similarly activated T blasts from BALB/c, DBA/1 or DBA/2 mice. It also reacts with COS cells transfected with mFASL cDNA derived from C57BL/6 and C3H mice, but not with mFASL cDNA from BALB/c nor DBA/2 mice. In addition, this clone efficiently blocks cytotoxic activity of a C3H T cell line but not a BALB/c T cell line. Phenotypic and genotypic characterizations of mouse FAS-Ligand reveal the existence on activated T cell lines and in spleen, testis and eye. FASL mRNA has been demonstrated at various levels in bone marrow, thymus, spleen, lymph node, lung, small intestine, testis and uterus. Morever, T cell activators but not B-cell activators enhanced the expression of FasL mRNA in splenocytes; and FasL mRNA was restricted to the T cell lineage among a panel of cell lines from lymphoid tissues. FasL is not functional in mice homozygous for the gld (generalized lymphoproliferative disease) mutation; these mice cannot limit the expansion of activated lymphocytes and develop autoimmune disease. FasL is a type II membrane protein a member of the TNF/NGF family, which binds to CD95 (FAS). FasL and its counter-receptor CD95 are believed to participate in T cell development, the regulation of immune responses, and cell mediated cytotoxic mechanisms. Functional studies suggest that mFasL.2 has higher specific activity than mFasL1. It has been demonstrated that human FASL is released from the surface of transfectant cells and activated human T cells by metalloproteinases.

Application: -immunofluorescent staining in the presence of metalloproteinase inhibitor on mFAS-L transfected and activated T cell lines (1-4ug/million cells). NOTE:PE has been reported to bind to mouse B cells via FCgII receptors. For immunostaining of mouse cells, this can be prevented by preincubating cells with a CD16/CD32 antibody.

DATA SHEET:    Hamster anti-MOUSE CD95L FAS-L    clone MFL3

Catalog#: RDI-MCD95L-FL3    $500.00/vial

(no azide formulation, cat#RDI-MCD95L-FL3X     $750.00/0.5mg

Biotin conjugated cat#RDI-MCD95L-FLBT    $625.00/0.5mg

Package Size: -500ug purified in 50mM Tris-Hcl (pH 8.0), 0.15M NaCl and 0.1% sodium azide.

Prep: purified from tissue culture supernatant via affinity chromatography.

Species: Armenian Hamster IgG

CLONE: MFL3

Antigen: C57BL/6 mouse FasL c DNA transfected baby hamster kidney cells

Activity: Clone MFL3 reacts with mouse Fas Ligand (Fas L), a member of the TNF/NGF superfmaily. Fas L has been reported to be expressed by activated mouse T cells as well as some T-cell lines and clones. Mouse FasL mRNA has been found in spleen, thymus, lung, lymph nodes, eye and testis. Fas L is a type II transmembrane protein which binds to its counter receptor CD95 (FAS) and is speculated to participate in regulation of a variety of immunological responses and lymphocyte development. Clone MFL3 has been reported to efficiently inhibit the cytotoxicity of mouse FAS-L transfected cells against human Fas-transfected cells.

Note: Because FAS-L is expressed at low density on activated cell, we recommend using a biotin labelled secondary antibody (please inquire) along with Streptavidin:PE
