Background: Vitamin D3 receptor (VDR) has been found in the intestine, bone
and kidney as well as skin and many cells of the endocrine and immune systems.
VDR is a member of the superfamily of steroid hormone receptors which includes
estrogen, progesterone, glucorticoid, androgen, and thyroid hormone receptors.
Vitamin D is known to play a role in calcium and phosphorous homeostasis,
bone remodeling, and regulation of cellular growth and differentiation.

antigen: partially purified chicken intestinal cytoplasmic 1,25 (OH)2D3 receptor protein. Hybridoma formed by fusing splenic lymphocytes derived from these Lewis rats with the myeloma cell line SP2/0-Ag14.
Specificity: -recognizes with high affinity just C-terminal to the DNA binding domain of the VDR; studies suggest the antigenic site lies between amino acids 89 to 105 of human VDR. It reacts with both the occupied and unoccupied forms of the receptor, and binds to VDR from a variety of tissues and cell types.
-recognizes VDR in the intestinal tissue of fish, frog, chick and rat, as
well as kidney from rat. It has also shown reactivity with VDR in cultured
cells from human breast, human intestine, rat bone, hamster ovary, pig kidney,
and mouse fibroblast. It has shown no reactivity with estrogen and glucocorticoid
Use: -Western blot:1ug/ml starting dilution detects the band approx 52-55kda in mammalian species, but band is approx 58kDa and 60Kda in avian.
-Histochemistry: Bouin's fixative recommended. 5ug/ml (strong nuclear staining)
-immunoprecipitation (titer must be determined)
Storage: Store at -20 DEG C upon receipt. Avoid frequent freeze thaw cycles. Recommend aliquoting. BEFORE OPENING, suggest centrifuging for 1 minute at 1000 RPM to concentrate material in vial.

This Pab has been successfully used in Western blot, immunoprecipitation and gel shift procedures. By Western blot, this antibody detects an ~53 kDa protein representing VDR in COS-7 cells transfected with the human gene. It supershifts DNA fragments that contain VDR response elements (e.g., rat osteocalcin and mouse osteopontin upstream elements).

Background: Steroid receptors are ligand-dependent intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate hormone. The 1,25-dihydroxy-vitamin D3 receptor (VDR) belongs to the superfamily of steroid hormone receptors which includes estrogen, progesterone, glucocorticoid, androgen, and thyroid hormone receptors. Vitamin D modulates calcium and phosphorus homeostasis, bone remodeling, cell growth regulation, and differentiation. Studies have found VDR in the intestine, bone, kidney, epidermis, and cells of the endocrine immune system. Repression of T-cell proliferation and interleukin-2 (IL-2) gene expresion, for example, occurs when VDR binds within the IL-2 enhancer. Formation of VDR/retinoic X receptor (RXR) heterodimers in the presence of intracellular 1,25(OH)2D3 has been shown to interfere with assembly of nuclear factor of activated T-cells (NFATp)/Fos/Jun/DNA complex and subsequent IL-2 gene transcription.

Vitamin D3 Receptor (VDR) is a 48 kDa hormone-inducible transcription factor that acts in the regulation of a number of cellular functions. PanVera's Vitamin D3 Receptor is both soluble and functionally active, and tested for its ability to bind [3H]1 alpha,25-dihydroxyvitamin D3. Like other members of the steroid hormone receptor superfamily, VDR binds to appropriate response elements upstream of the regulatable gene. For example, VDR binding to a site upstream of the osteocalcin gene regulates calcium homeostasis.
