Applications: -Immunohistochemistry:1:5K-1:10K using 24hour incubation on formalin,
Bouin's or Zamboni's fixed tissue. Will react with paraffin embedded tissue.
-Immunocytochemistry: 1:1K-1:200K (24 hour incubation on fetal rat neurons_
-Immunoblotting: 1:1K

vasopressin (AVP) exerts its action through binding to specific membrane receptors coupled to distinct second messengers. There are three types of AVP receptors: V1a, V1b and V2 subtypes. The V2 receptor stimulates adenyl cyclase and protein Kinase A, V1 activates phospholipase A2.C and D, resulting in prodcution of IP3 and DAG, the mobilization of intracellular calcium, the influx of extracellular calcium, the activation of protein kinase C, and protein phosphorylation. The V1a receptors mediate vasoconstriction and hepatic gluconeogenesis platelet aggregation, coagluation factor release. V1a receptors are found in vascular smooth muscle, hepatocytes, blood platelets, lymphocytes and monocyte, tyep II pneumocytes, adrenal cortex, brain, reproductive organs, retinal epithelium, renal mesangial cells.
the V1b modulates ACTH secretion from the naterior pituitaary and the V2 mediates anti-diuretic effects. V1b receptors are located in anterior pituitary. V2 renal receptors are present in medullary portion of the kidney where they control free water and urea absorption by stimulating adenyl cyclase.
AVP receptors are members of the G protein coupled receptors with putative 7 transmembrane domains. the sizes of various AVP receptors are V1a (rat, 424aa, human 418aa); V1b(rat, 421aa, human 424aa); V2 (rat, 371aa, human 371aa). The N terminus and C-terminus are predicted to be extracellular and cytoplasmic, respectively.

Immunogen: an 18 amino acid peptide between the 5th and 6th transmembrane
domain of rat AVP-V2 (predicted to be the cytoplasmic domain)
specificty: reacts with the arginine vasopressin receptor (AVP-2) in rat. The peptide used for immunization has no significant simililarity with AVP V1a or V1B.
species reactivity: rat (not tested with other species) by antigen homology
(human and mouse expected)
Applications: -western blot:1-10ug/ml with chemiluminescence detection, approx
62kda band in mebranes from rat inner medualla. N-Gluconase redcued the size to
approx 36 kda (deglycosylated form)
-histochemistry: 2-10ug/ml on pfa fixed tissue. AVP-V2 has been localized in terminal IMCD and other areas of rat kidney.
-Optimum titer must be established by the end user

-84% homology with mouse (16/19aa)
-68% homology human 13/19aa
-52% homology with sheep (10/19aa)
