Monoclonal antibody: 5H9, 10E1, 10E2, 4G11, 10A5, 3C8, 10H9, 18A10, 11E4
Hybridoma clones, producing MAbs, have been derived from hybridization of SP2/0 myeloma cells with spleen cells of Balb/c mice immunized with purified human PAPP-A antigen.
Specificity: Human PAPP-A antigen purified from placental blood (heterotetrameric complex, consisting of two PAPP-A subunits and two pro-MBP subunits) Clone 11E4 is specific to proMBP subunit.
Mouse isotype: IgG2b for MAb 5H9, 10E1 and 10E2
IgG2a for clones 4G11, 10A5, 3C8, 10H9 and 18A10
Application: Detection of human PAPP-A antigen in immunoassay.
Recommended pairs (coating - conjugate) : for the detection of PAPP-A in blood of AMI patients and patients with unstable angina (capture - detection): 4G11 – 3C8, 4G11 – 10H9, 10E2 – 10E1
Best pairs for the detection of PAPP-A in pregnancy serum (capture – detection): 4G11 – 3C8, 4G11 – 10H9, 10A5 – 3C8, 10E2 – 10E1, 10E2 – 5H9, 5H9 – 10E2
MAbs 4G11, 10A5, 18A10 (best) and 10E1 recognize PAPP-A subunit in the heterotetrameric complex in Western Blotting. MAb 11E4 recognizes proMBP subunit in Western Blotting
Purification: All MAbs are purified by chromatography on protein A Sepharose. Clone 10E2 is additionally purified by gel-filtration. Purity of all MAbs is tested by electrophoresis.
Presentation: Each vial contains antibodies in PBS, pH 7,4, containing 0,1% of sodium azide as preservative. Store at + 4 °C.
Material safety note: This product is sold as an antibody preparation for research purpose only. Standard Laboratory Practices should be followed when handling this material. Contains sodium azide (0,1%) as preservative. Although the amount of sodium azide is very small appropriate care must be taken when handling this product.
