-immunogen: tyrosine hydroxylase purified from PC12 rat pheochromocytoma cells

-reacts with rat tyrosine hydroxlase. It is useful for identifying tyrosine hydroxylase containing catecholaminergic neurons in rat brain

-suitable for immunohistochemical staining of frozen tissue sections for use in light and electrom microscopy (frozen tissue sections of rat brain)

Mouse anti-TRH, TYH & PAH  (Tryptophan, Tyrosine and Phenylanaine Hydroxlases)

-immunogen: monkey liver phenylalanine hydroxylse

-reacts with a common epitope of TRH, TYH and PAH. The epitope has been localized to the human PAH sequence, rat TYH, and rabbit TRH, with 11 out of 17 amino acids bing identical. PH8 reacts with human, rat, monkey, and rabbit. Additionally, PH8 has been shown to rewact with PAH in western blots of liver extracts from a wide range of other species including dog, pig, ox, koala, chicken, trout and toad, and with TYH in dog frozen tissue sections. Clone PH8 ab did not demonstrate cross-reactivity to PAH in western blots of squid liver extracts (despite the presence of PAH activity)

-Application Note: Clone PH8 ab is particularly useful in analyzing neuronal distribution by immunohistochemical staining:in frozen tissue sections, PH8 binds to TYH and TRH and can thus identify TYH-containing catecholamergic neurons and TRH- containing serotonergic neurons. Cell types in frozen tissue can be distinguished by variation in staining intensity. At higher antibody concentrations (>0.25ug/ml), all monoaminergic neurons are stained without distinction between serotonergic and catecholaminergic cells. At lower antibody concentrations (approx 0.1-0.25ug/ml) only serotonergic cells will stain.

-in formalin fixed paraffin embedded tissue sections, PH8 binds to TRH but not TYH, possible due to a change in the antigenic determinant caused by fixation. Thus, PH8 can be used to specifically identify serotonergic neurons in fixed tissue. This is particulalry notable since serotonin is rapidly metabolized and thus undetectable by serotonin-specific antibodies in post- mortem tissue. TRH immunoprecipiatted with PH8 retains substantial enzymatic activity.

-PH8 can also be used for immunoprecipitation and western blot analysis of TRH. PH8 is not recommended for immunoprecipitation or western blot analysis of TYH.

-for PAH, clone PH8 ab is also useful for immunohistochemical staining of frozen and paraffin-embedded liver tissue sections. It can also be used for immunoprecipitation and western blot analysis of PAH. PH8 ab does not substantially inhibit activity of purified PAH.
