Antigen: SV40 transformed Balb/c mouse fibroblasts

specs: recognizes an epitope between aa 270-520 of SV40 large T antigen. Large T is a multifunctional 85kD phosphoprotein that is the sole viral protein required for Sv40 replication, and also causes transformation of susceptible cell lines. Studies have suggested that clone Pab100 binds preferentially to newly synthesized large T, and to ATPase active large   T. In other experiments, it appears to bind preferentially to mature large T.

                       -indirect immunofluorescence of tissue cultured cells

                       -histochemistry of formalin fixed paraffin embedded tissue sections -

                      -ATPase-active large T immunoprecipitated by PAB100 has been shown to retain ATPase activity

specs: recognizes a C terminal epitope within the last 190 amino acids. Studies suggest that Pab101 binds the strongest to mature T-ag.

Applications: -immunoprecipitation (1-2ug/million cells), T-ag immunoprecipitated as a single or multiple bands between 80-95kda depending on post-translational modification.

Background: Simian virus SV40 has provided an important model for studies of cellular mechanisms involved in a malignant transformation. The major SV40 translational products include the 94 kDa large T antigen and the 21 kDa small T antigen, both of which are encoded by the early region of the SV40 viral genome. The large T antigen complexes with the p53 suppressor gene resulting in its functional inactivation and thus promoting cell transformation. In addition SV40 large T antigen binds DNA polymerase and the transcription factor, AP-2 and forms complexes with a second tumor supressor gene-encoded protein, Rb 105. Binding of SV40 T antigen is specific for the "pocket" domain of Rb p105 which is also the binding site for the E2F cellular transcription factor.

Clone: PAB416-derived by immunization with purified large T antigen and fusion of mouse spleen cells with NS-1 mouse myeloma cells.

Isotype: mIgG2a

Specificty: Clone PAB416 Mab SV40 T Ag is reactive by immunoprecipitation with the 94,000 dalton SV40 large T antigen (1) and stains SV40 infected cells. The epitope is between Ile 83 and Lys 128 of the SV40 large T antigen (11). -specific for antigenic determinants unique to the SV40 large T antigen and non-reactive with SV40 small T antigen (1)

APPLICATIONS: -western blot: 1-2ug/ml

                             -immunofluorescence:1-5ug/ml

                              -immunoprecipitation:

                              SV-T2 and SV80 cells can be used as a positive control and 3T3 cells can be used as a negative

                              control.

BACKGROUND: SV40 T Ag is a mouse monoclonal antibody with . Both antigens are encoded by the early region of the SV40 genome (2). The large T antigen binds DNA (3), and complexes with a 53,000 dalton cellular transforming protein, p53 (4-6), which is required for initiation of viral DNA replication during lytic growth (7). In addition the l arge T antigen binds DNA polymerase and the transcription factor AP-2 (8) and forms a specific complex with the p105 product of the retinoblastoma susceptibility gene (9 and 10).

Specificty: Clone PAB419 Mab SV40 T Ag is reactive by immunoprecipitation with the 94,000 dalton SV40 large T antigen and the 20,500 dalton SV40 small T antigen (1) and stains SV40 infected cells. The epitope is in the amino terminal domain of SV40 large T antigen (also shared with SV40 small T antigen)

Background: SV40 T Ag clone Pab419 is a mouse monoclonal antibody with specificity for antigenic determinants localized to the amino terminal domain of the SV40 large T antigen (1). The sequences recognized by this reagent are also present on SV40 small T antigen (1). Both antigens are encoded by the early region of the SV40 genome (2). The large T antigen binds DNA (3), and complexes with a 53,000 dalton cellular protein, p53 (4), which is required for initiation of viral DNA replication during lytic growth (5). In addition the large T antigen binds DNA polymerase and the transcription factor AP-2 (6) and forms a specific complex with the P105 product of the retinoblastoma susceptibility gene (7 and 8).
