RDI Divison of Fitzgerald Industries Intl offers a wide line of antibodies. Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Please inquire for other applications or types of antibodies not listed below.

Presentation: 100ug affinity pure antibody (lyophilized from 0.1ml (1mg/ml) 100mM Tris pH7.5 with 0.2% BSA and 0.05% sodium azide).
Reconstitution: Add 100ul sterile DI water, let set at least 30 minutes at 4 DEG C. Getly mix well. If desired, centrifuge 1-2 minutes at 1000 rpm to concentrate material in vial. Recommend making dilutions in buffers containing 0.1-1% BSA and 0.05% sodium azide.
Purification: peptide affinity purified.
Antigen: A 15 amino acid peptide sequence between transmembrane domain 7 and 8 of rat serotonin transporter (SERT, also know as 5-HT transporter) was used to generate anti-rat SERT This region is presumably from the extracellular domain. A cysteine has been added at the NH2 terminus for coupling to KLH
Reconstitution: All peptide/antibody received in solution or reconstituted from Lyophilized vials should be stored frozen at -20oC or below in suitable aliquots. It is not recommended to store diluted solutions. Avoid repeated freeze and thaw.
Specificity: the 15 aa rat SERT peptide sequence is 100% homologous in human ,mouse and monkey and 93% in guinea pig.
USE: Western Blotting (1-10 ug/ml for affinity pure using Chemiluminescence technique). An antibody raised against a similar peptide has been shown to react with a SERT in baculovirus cells expressing recombinant rat and human-SERT. Antibody activity may be reduced considerably when detecting endogenous SERT from rat or human cells. SERT has been observed to migrate as 94 kDa (rat platelet),80 kDa (pulmonary membranes, and 76 continued kDa (rat brain). This could be due to differences in postranslational modifications (e.g.,glycosylation).
ELISA (1:100K; using 50-100 ng SERT control peptide/well).
Immunoprecipitations . An antibody to the SERT epitope has been shown to be effective in immunoprecipitating the 61 kDa species from rat and human SERT-transfected HeLa cells. approx 1-5ug/mg protein
Histochemistry & Immunofluorescence: Not tested. We recommend the testing of affinity purified antibody at 10-50 ug/ml on frozen sections of tissues fixed with 4% paraformaldehyde.

Control peptides are lyophilized from 1 mg/ml solution (100 g/100 l) in PBS + 0.05% sodium azide. Reconstitute with 100ul DI water, let set at least 30 minutes, mix gently. Generally titer antibody to maximum usable dilution and then pretreat antibody with 5-10X weight of peptide for 2-3 hours RT or overnight 4 DEG C, then proceed with staining. -Control peptides are small peptides and cannot be used to run on westernblot.

Specificity: Binds to rat and mouse serotonin transporter
Immunogen: rat serotonin transporter, N-terminus/GT fusion protein (aa 1-85)
Application: Immunohistochemistry:approx 1ug/ml on PFA fixed brain and adrenal tissue. (not tested on paraffin sections) -not recommended for western blot

Storage/Handling: The undiluted antibody solution is stable for approximately 6 months when stored between 2 and 8'C. For longer storage, store at -20'C. Avoid frequent freeze thaw cycles.

Important Note: We recommend gently tapping the product vial on a hard surface or briefly spinning the vial in a tabletop centrifuge to dislodge and liquid entrapped in the container's cap during shipment.
