Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Storage: Antibodies are stable for six months from date of receipt when stored at -20 ° C to -70 ° C.The antibody can be stored at 2 ° -8 ° C for 1 month without detectable loss of activity.Upon receipt, antibody should be aliquotted and stored frozen at -20 ° C to -70 ° C in a manual defrost freezer. Avoid repeated freeze-thaw cycles.

Notes: Histochemistry: Antiserum was used on perfusion   fixed tissue. Perfusion: paraformaldehye-picric acid fixative, and 3) 10% sucrose in PBS as a cryo-protectant. Desired tissues were dissected and stored overnight in 10% sucrose in PBS. Slide-mounted tissue sections were processed for indirect immunofluorescence. Slides were incubated with blocking buffer for 1 hour at room temperature. Primary antiserum was diluted with blocking buffer to the appropriate working concentration. Blocking buffer was removed and slides were incubated for 18-24 hours at 4 ºC with primary antiserum. Slides were rinsed 3 times and then incubated with secondary antibodies for 1 hour at room temperature. Slides were again rinsed 3 times and coverslipped. Staining was examined using fluorescence microscopy.

Note: Sodium azide (NaN3) interferes with peroxidase reactions and should not be used with peroxidase methodologies. If sodium azide is present in any steps of the staining procedure, the tissue should thoroughly be rinsed with sodium azide-free buffer before performing the peroxidase reaction.

DATA SHEET : Rabbit anti-Human Serotonin (5-HT)   (Paraffin Sections)

Preparation: Lyophilized antibody delipizied from rabbit serum diluted in 1% w/v BSA in PBS with 15mM sodium azide. Reconstitute with 1ml of sterile distilled water. Let set at least 30 minutes, mix well.

Antigen: Serotonin creatinine sulfate complex conjugated to BSA

Specificity: human serotonin (5-hydroxytryptamine, 5-HT) other species not tested

USE: Effective on paraffin wax embedded tissue (not tested on frozen)

Protocol: Immunohistochemistry:Typical working dilution 1:100 - 1:200. 60 Minutes incubation at 25'C. Standard ABC technique.

Positive controls: small intestine

Staining pattern: cytoplasmic

Storage and stability: Store unopened lyophilized antibody at 4'C. Under these conditions, there is no significant loss in product performance up to the expiry date indicated on the vial label. The reconstituted antibody is stable for at least two months when stored at 4'C. For long term storage, aliquot in non frost freeze freezer, Avoid repeated freeze thaw cycles. Background:Serotonin is a widely distributed neurotransmitter and hormone in the mammalian central nervous system (CNS) and periphery. Serotonin is formed by the decarboxylation of 5-hydroxy-tryptophan, its intermediate, which in turn is formed by hydroxylation of L-tryptophan hydroxylase. In the CNS, the action of 5HT is terminated by reuptake into the presynaptic terminal by specific 5-HT transporters. 5-HT has been implicated in several neuropsychiatric disorders such as anxiety, depression and schizophrenia. The majority of serotonergic nerve terminals in the CNS originate in neuronal cell bodies of the rephe nuclei (dorsal, median), nucleus raphe obscurus and nucleus raphe pallidus in the brainstem which project to specific areas of the brain and spinal cord. 5-HT is thought to be an inhibitory neurotransmitter regulating a wide range of sensory, motor and cortical functions in the CNS. In the periphery, 5-HT is present in neural and non-neural structures such as platelets, gastrintestinal tract (myenteric plexus, enterochromaffin cells), lungs (neuroepithelial cells) thyroid gland and spleen. This antibody s useful for the detection in normal GI tract, appendix and in carcinoid tumours arising from the gut.

Positive  Control: Appendix

Localization: Cytoplasmic, reacts with serotonin-containing cells in a wide range of normal, reactive, and neoplastic tissues. Other endocrine cells, such as those of pancreatic islets and pituitary gland, are negative with this antibody.

Storage: Store vial at 4°C.

Descript on: Serotonin-positive cells are present in gastric, duodenal, jejunal, ileal, colonic, and appendiceal mucosae. Positive cells are sparse and usually occur in the lower half of the gastric and intestinal mucosae. In the appendix, in addition to the mucosal serotonin-containing cells, there are single or small clusters of serotonin-positive cells in the sub-epithelial region of the lamina propria.
