Mouse anti-Replication Protein A (RPA)p32

Immunogen: purified recombinant human RPA protein

Clone: 9H8 (also referred to as alphaSSB34A)

Reactivity: human and mouse (others not tested)

Uses: -Western blot: approx 1:250 dilution

         -Gel supershift

         -immunoprecipitation: approx 10ul/mg protein lysate

        -blocks SV40 DNA replication in vivo

        -no effect on Binding of RPA to ssDNA

        -blocks stimualiton of DNA Pol alpha

Positive Control: HeLa cells or HEp-2 cells

Localization: nuclear

Background: Recognizes a protein of 34kDa, identified as a subunit of Replication Protein A (RPA), also known as DNA Binding   protein or HSSB). RPA from human cells is a stable heterodimer of 70kDa, 32-34kDa and 11-14Kda subunits (RPA70, RPA32 and RPA14 respectiviely). It is involved in DNA replication, repair and recombination. RPA interacts with multiple proteins, including proteins required for the initiation of SV40 DNA replication, DNA polymerase alpha and SV40 large T antigen. The DNA binding activity of human RPA is localized to the residues 181-422 of the 70Kda subunit. RPA34 is phosphorylated at the G1/S boundary of the cell cycle or upon exposure of cells to DNA damaga-inducing agents including ionizing and Uv radiation. Phosphorylation of ZRPA may play a role in coordinating DNA metabolism in the cell. RPA may also have a role in modulating gene expression. RPA and the human repair protein XPA specifically interact both in vitro and in vivo.
