Clinical Application/ Specificity Antibody recognizes desmoglein 2 present in all desmosome possessing tissues; stratified and single-layered polar epithelia and tumors derived therefrom as well as heart muscle and dendritic reticulum cells of lymphatic follicles.
With stratified squamous epithelia only desmosomes connecting the basal, proliferative cell layer contain detectable desmoglein 2, it seems to be absent from all suprabasal cell layers (these layers are, however positive for desmoglein 1 and 3). In basal epithelial cells,desmoglein 2 expression is generally very low.
Application o Western blot technique
o Immunohistochemistry on cryostat* and paraffin** sections
o ELISA
* for better resolution on frozen sections preincubation (direclty after fixation) with 0.05-0.2% Triton X-100, for 5-10 minutes, depending on tissue type, is recommended (see ref#3)
