1.0 mg/ml stock in PBS (pH7.2) with 1% sucrose & 0.1% Sodium azide. *When disposing azide into
a drain, flush with plenty of water.

Isotype: Mouse IgG1
Derivation: purified from mouse ascites fluid using
protein-A Sepharose. This hybridoma
was established by fusion of mouse myeloma P3U1 with Balb/c
splenocyte immunized with -human
brain GAD 65 fusion protein corresponding to full length amino acids (1-585 a.a.).
SPECIFICITY : reacts with GAD 65 and GAD 67 by Western blotting (approx 1:1000) and immunohistochemistry (1- 10ug/ml) staining. This antibody detects 65KD and 67KD of human and mouse GAD by Western blotting on total cell lysate from mouse brain. Not tested with other species -Positive control include human and mouse brain lysates.
STORAGE : Store lyophilzed antibody at 4 DEG C on arrival. After reconstitution, store 4 DEG C short term or recommend aliquoting and store at -20 DEG C in non-frost free freezers. Avoid frequent freeze thaw cycles.
BACKGROUND : Glutamic acid decarboxylase (GAD) catalyzes formation of gamma -Aminobutyric acid (GABA), which is the inhibitory neurotransmitter, and is present in brain as well as several tissues outside the central nervous system. Biological functions of GAD and GABA extend beyond regulation of neurotransmission to include effects on the immune system as well as modulation of cell proliferation, protein synthesis, and metabolism 1) . Two isoforms of GAD, GAD 65 and GAD 67 each derives from a single separate gene, were isolated from human fetal brain lambda gt11 cDNA library 2)

Background: gamma-aminobutyric acid (GABA) is an amino acid neurotransmitter that is considered to be the major inhibitory neurotransmitter in the mammalian central nervous system. The highest concentrations of GABA are found in the brain, where it is synthesized from glutamic acid to GABA by an enzyme called glutamic acid decarboxylase (GAD). GAD is also expressed in the insulin-producing â cells of the islets of Langerhans. Two isoforms of GAD are present in rat brain, GAD65 and GAD67, based on their relative molecular weight in kDa. Both isoforms have significant levels of homology in the catalytic portion of the molecule, but differ greatly in the first 95 amino acids in the N-terminal region. GAD65 migrates at ~65 kDa in SDS/PAGE.
