Reactivity: human, mouse, pig and bovine(others not tested)

Uses: -Western blot: 1ug/ml for 2 hours rt (use immobilon membranes)

        -histochemistry (acetone fixed frozen sections, alcohol fixed paraffin, not with routine formalin fixed paraffin approx 10ug/ml for 60 minutes RT

        -Immunohistology (Frozen and Acid-alcohol-fixed, paraffin-embedded 1 ) (Use Ab at 2µg/ml for 30 min at RT)

* Staining of acid-alcohol fixed tissues requires antigen unmasking with 15,000U/ml of bovine testicular hyaluronidase (Sigma) in PBS, pH 7.4 for 30 min at 37C

Positive Ctrl: breast carcinomas, squamouse cell carcinomas

Localization: basement membrane

Background: Perlecan is a major heparan-sulfate proteoglycan (HSPG) within all basement membranes and cell surfaces. Because of its strategic location and ability to store and protect growth factors, perlecan has been strongly implicated in the control of tumor cell growth and metastatic behavior. Perlecan possesses angiogenic and growth-promoting attributes primarily by acting as a coreceptor for basic fibroblast growth factor (FGF-2). Suppression of perlecan causes substantial inhibition of neoplastic growth and neovascularization. Thus, perlecan is a potent inducer of neoplasm growth and angiogenesis in vivo and therapeutic interventions targeting this key modulator of tumor progression may improve neoplastic treatment. This antibody does not cross-react with (laminin, dermatran sulfate proteoglycan) or fibronectin.
