Background: Paxillin is a focal adhesion protein and a substrate for several tyrosine kinases such as src, FAK, and p120BRC/ABL. The tyrosine phosphorylation of paxillin is affected by conditions that change cell-cell adhesion, consistent with the belief that paxillin is involved in the regulation of cell morphology. Additionally because of is SH3-binding domain, paxillin associates tightly with FAK and Crk in a manner that is independent of the extracellular matrix. Although paxillin was initially detected in fibroblasts, its phosphorylation may also be important during neurite extension during differentiation.

Antigen: derived from chick paxillin

package: 50ug (200ul) or 150ug (600ul) purified antibody (0.25mg/ml) in a 50%glycerol, 20mM sodium phosphate pH7.5, 150mM NaCl, 1mg/ml BSA and 1.5mM sodium azide.

Use: Suitable for western blot on human, rat, mouse, dog and chick using approx 0.5 ug/ml ab on total cell lysate using human endothelial cells as positive control. Major band at Mw 68kDa. Recommend 2-3 hour RT or overnight 4 DEG C incubation with enhanced chemiluminescence detection. Also suitable for immunofluorescence.

Storage: Store at -20 DEG C upon receipt. Avoid frequent freeze thaw cycles. Recommend aliquoting. Avoid frequent freeze thaw cycles.

-for paraffin sections, requires boiling tissue sections in 1mM EDTA pH8.0 for 10-20 minutes followed by cooling at RT for 20 minutes.
